Abstract

Official abstract text for this publication.

The invention provides peptides and the nucleic acid sequences that encode them. The invention further provides therapeutic, diagnostic and research methods for diagnosis, treatment, and prevention of apoptosis associated disorders.

First claim

What is claimed is: 1. A method of screening for an apoptotic sensitizer compound comprising: (a) contacting mitochondria from a cell that overexpresses BCL-2 or BCL-XL with an isolated BH3 peptide comprising SEQ ID NO: 1 to form a BCL-2-peptide complex or a BCL-XL-peptide complex, wherein the BH3 peptide is less than or equal to 35 amino acids in length, and wherein the BH3 peptide when displaced from the complex activates pro-apoptotic BAK to thereby induce BAK oligomerization and cytochrome c release; (b) contacting said complex with a candidate apoptotic sensitizer compound that does not activate pro-apoptotic BAK; and (c) determining cytochrome c release from said mitochondria, wherein an increase in the amount of cytochrome c release in the presence of said candidate apoptotic sensitizer compound compared to the amount of cytochrome c release in the absence of said compound indicates said compound is an apoptotic sensitizer compound that displaces the BH3 peptide from the BCL-2-peptide complex or the BCL-XL-peptide complex. 2. The method of claim 1 , wherein the BH3 peptide is less than or equal to 25 amino acids in length. 3. The method of claim 1 , wherein the BH3 peptide consists of SEQ ID NO:1.
