Provided herein, inter alia, are fusion DNA constructs comprising improved protease recognition sequences for expressing and purifying one or more polypeptides of interest as well as methods for producing one or more polypeptides of interest in a recombinant host cell.

We claim: 1 . A fusion polypeptide comprising a KEX2 region, wherein the amino acid sequence of the KEX region comprises TKVAVEKRQV (SEQ ID NO:10), wherein the amino acid sequence is completely cleaved by one or more protease(s) and wherein the protease is a Kex2 serine peptidase (EC 3.4.21.61). 2 . The fusion polypeptide of claim 1 , further comprising a polypeptide encoding a signal sequence. 3 . The fusion polypeptide of claim 2 , wherein the polypeptide encoding a signal sequence is located N-terminal or C-terminal from the amino acid sequence of the KEX region. 4 . The fusion polypeptide of claim 1 , further comprising a polypeptide encoding a carrier protein. 5 . The fusion polypeptide of claim 4 , wherein the polypeptide encoding a carrier protein is located N-terminal or C-terminal from the amino acid sequence of the KEX region. 6 . The fusion polypeptide of claim 5 , wherein the polypeptide encoding a carrier protein is adjacent to the polypeptide encoding a signal sequence. 7 . The fusion polypeptide of claim 5 , wherein the carrier protein comprises CBH1 or a fragment thereof.
