Provided herein are Itraconazole analogs. Also provided herein are methods of inhibition of Hedgehog pathway, vascular endothelial growth factor receptor 2 (VEGFR2) glycosylation, angiogenesis and treatment of disease with Itraconazole analogs.

What is claimed is: 1 . A method of treating a cancer in a subject in need thereof, comprising administering to the subject an effective amount of a compound selected from: Compound Structure Number 208 214 215 or an optically pure stereoisomer or pharmaceutically acceptable salt thereof, wherein the cancer is selected from prostate cancer, non-small cell lung cancer, and basal cell carcinoma. 2 . The method of claim 1 , wherein when the cancer is prostate cancer or non-small cell lung cancer, the compound is administered via oral or parenteral administration; and wherein when the cancer is basal cell carcinoma, the compound is administered via oral, parenteral, or topical administration.
