Disclosed are a method and a reagent for quantifying HDL3 in a test sample without requiring laborious operations. The method for quantifying cholesterol in high-density lipoprotein 3 comprises reacting a test sample with one or more surfactants which react specifically with high-density lipoprotein 3, and quantifying cholesterol. When one surfactant is used, the surfactant is one selected from the group consisting of polyoxyethylene polycyclic phenyl ethers having an HLB of 12.5 to 15. When two or more surfactants are used, at least one of the surfactants is at least one selected from the group consisting of polyoxyethylene polycyclic phenyl ethers, and the two or more surfactants are combined so as to provide the total HLB of 12.5 to 15 of the combined surfactants.

The invention claimed is: 1. A reagent for quantifying cholesterol in high-density lipoprotein 3 in a serum, said reagent comprising two or more surfactants which react specifically with high-density lipoprotein 3, and cholesterol oxidase and cholesterol esterase; wherein said two or more surfactants are selected from the group consisting of polyoxyethylene styrenated phenylethers, and the two or more surfactants are combined so as to provide the total HLB of 13.7 to 14.0 of the combined surfactants; wherein a total concentration of said surfactants is 0.25 to 1.25 w/v % in terms of the final concentrations; and wherein said two or more surfactants provide a reaction ratio of serum high-density lipoprotein 3 to high-density lipoprotein 2 (HDL3/HDL2) of 4.6 to 10.6. 2. The reagent according to claim 1 , wherein said two or more surfactants are selected from the group consisting of polyoxyethylene distyrenated phenylethers. 3. The reagent according to claim 1 , wherein the total HLB of the used surfactants is 13.8 to 14.0. 4. The reagent according to claim 1 , wherein the total HLB of the used surfactants is 13.9 to 14.0. 5. The reagent according to claim 1 , wherein the reagent contains Sphingomyelinase. 6. The reagent according to claim 1 , wherein a total concentration of said surfactants is 0.25 to 1.0 w/v % in terms of the final concentrations. 7. The reagent according to claim 1 , wherein a total concentration of said surfactants is 0.25 to 0.75 w/v % in terms of the final concentrations. 8. The reagent according to claim 1 , wherein said two or more surfactants provide a reaction ratio of high-density lipoprotein 3 to high-density lipoprotein 2 (HDL3/HDL2) of 6.6 to 10.6.
