This disclosure relates to an anti- Pseudomonas Psl binding molecule and uses thereof, in particular in prevention and treatment of Pseudomonas infection. Furthermore, the disclosure provides compositions and methods for preventing and treating Pseudomonas infection.

What is claimed is: 1. An isolated polynucleotide comprising a nucleic acid encoding an antibody or antigen-binding fragment thereof, wherein the antibody or antigen-binding fragment comprises a heavy chain variable region (VH), wherein the VH comprises a VH complementarity determining region-1 (VHCDR1), VDCDR2, and VHCDR3 of amino acid sequences SEQ ID NO:47, 48, and 75, respectively, and the antibody or antigen-binding fragment comprises a light chain variable region (VL), wherein the VL comprises a VLCDR1, CDR2, and CDR3 of amino acids 50, 51, and 52, respectively. 2. A host cell comprising the polynucleotide of claim 1 . 3. The polynucleotide of claim 1 , wherein the antibody or fragment thereof comprises the VH and VL of SEQ ID NO:74 and SEQ ID NO:12, respectively. 4. A vector comprising the polynucleotide of claim 3 . 5. A host cell comprising the polynucleotide of claim 3 . 6. The polynucleotide of claim 3 , wherein the antibody or antigen-binding fragment thereof is a scFv. 7. A vector comprising the polynucleotide of claim 6 . 8. A host cell comprising the polynucleotide of claim 6 . 9. The polynucleotide of claim 1 wherein the antibody or fragment thereof is a monoclonal antibody, a humanized antibody, a chimeric antibody, a human antibody, an Fab fragment, an Fab′ fragment, an F(ab)2 fragment or a single chain Fv (scFv). 10. The polynucleotide of claim 9 , wherein the antibody or antigen-binding fragment thereof is a scFv. 11. A vector comprising the polynucleotide of claim 10 . 12. A host cell comprising the polynucleotide of claim 10 . 13. A vector comprising the polynucleotide of claim 1 . 14. The polynucleotide of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises a VH of SEQ ID NO:74. 15. A vector comprising the polynucleotide of claim 14 . 16. The polynucleotide of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises a VH of SEQ ID NO:12. 17. A vector comprising the polynucleotide of claim 16 . 18. An isolated polynucleotide comprising a nucleic acid encoding an antibody or antigen-binding fragment thereof that competitively inhibits Pseudomonas Psl binding by an antibody or antigen-binding fragment thereof comprising the heavy chain variable region (VH) and light chain variable region (VL) region of SEQ ID NO:74 and SEQ ID NO:12, respectively. 19. A vector comprising the polynucleotide of claim 18 . 20. A host cell comprising the polynucleotide of claim 18 . 21. A host cell comprising a polynucleotide encoding a VH comprising SEQ ID NO:74 and a polynucleotide encoding a VL comprising SEQ ID NO:12.
