Abstract

Official abstract text for this publication.

Anaplasma phagocytophilum surface proteins Asp14 and OmpA and homologous genes from Anaplasmatacaea family members are used in compositions suitable for vaccines to treat or prevent infections caused by tick-born bacteria of the Anaplasmatacaea family. Asp14 and/or OmpA proteins or peptide fragments may be used in combination with other Anaplasmatacaea surface proteins to elicit an immune response. Furthermore, antibodies to Asp14 and/or OmpA proteins can be used in diagnostic methods to determine whether an individual has contracted an Anaplasmatacaea infection. Because of the conserved invasin domains in the surface proteins, a wide range of Anaplasmatacaea infections may be diagnosed, treated or prevented using compositions of the invention.

First claim

What is claimed is: 1. An immunogenic composition comprising a combination of a first isolated polypeptide comprising SEQ ID NO: 03, a second isolated polypeptide comprising SEQ ID NO: 06, and an adjuvant in a vehicle or carrier suitable for administration to a subject. 2. The immunogenic composition of claim 1 , wherein one or more of said first and second polypeptides is linked to an amino acid spacer, an amino acid linker, a signal sequence, a stop transfer sequence, a transmembrane domain, a protein purification ligand, a heterologous protein, or one or more additional polypeptides comprising SEQ ID NO:01, 02, 03, 04, 05, 06, 07, 08, or 09. 3. The immunogenic composition of claim 2 , wherein said one or more polypeptides is linked to said protein purification ligand and said protein purification ligand is selected from the group consisting of GST and His. 4. An immunogenic composition comprising at least one fusion polypeptide comprising SEQ ID NO: 03 and SEQ ID NO: 06 in a vehicle or carrier suitable for administration to a subject. 5. The immunogenic composition of claim 4 , wherein one or more of said at least one fusion polypeptide is linked to an amino acid spacer, an amino acid linker, a signal sequence, a stop transfer sequence, a transmembrane domain, a protein purification ligand, a heterologous protein, or one or more additional polypeptides comprising SEQ ID NO: 01, 02, 03, 04, 05, 06, 07, 08, or 09. 6. The immunogenic composition of claim 5 , wherein said one or more polypeptides is linked to said protein purification ligand and said protein purification ligand is selected from the group consisting of GST and His. 7. An immunogenic composition comprising a combination of a first isolated polypeptide comprising SEQ ID NO:03 and a second isolated polypeptide comprising SEQ ID NO:06 in a vehicle or carrier suitable for administration to a subject, wherein one or more of said first and second polypeptides is linked to an amino acid spacer, an amino acid linker, a signal sequence, a stop transfer sequence, a transmembrane domain, a protein purification ligand, a heterologous protein, or one or more additional polypeptides comprising SEQ ID NO:01, 02, 03, 04, 05, 06, 07, 08, or 09. 8. A method of inducing an immune response in a subject comprising administering to said subject an immunogenic composition according to claim 1 . 9. The method according to claim 8 , wherein one or more of said first and second polypeptides is linked to an amino acid spacer, an amino acid linker, a signal sequence, a stop transfer sequence, a transmembrane domain, a protein purification ligand, a heterologous protein, or one or more additional polypeptides comprising SEQ ID NO:01, 02, 03, 04, 05, 06, 07, 08, or 09; or a combination thereof.
