Official abstract text for this publication.
The present invention provides implants comprising a therapeutic drug and a polymer containing polylactic acid (PLA) and optionally polyglycolic acid (PGA). The present invention also provides methods of maintaining a therapeutic level of a drug in a subject, releasing a therapeutic drug at a substantially linear rate, and treating schizophrenia and other diseases and disorders, utilizing implants of the present invention.

What is claimed is: 1. An implant comprising: a therapeutic drug and a polymer, said polymer comprising polylactic acid (PLA) and optionally polyglycolic acid (PGA) in a PLA:PGA molar ratio between 50:50 and 100:0, wherein said therapeutic drug is present in an amount of about 10%-60% (w/w), wherein the presence of said polymer in said implant extends the release duration of said therapeutic drug in a subject, and wherein said therapeutic drug is risperidone. 2. The implant of claim 1 , wherein said drug is present in an amount of about 30-60% (w/w). 3. The implant of claim 1 , wherein said drug is present in an amount of about 60% (w/w). 4. The implant of claim 1 , wherein said drug is present in an amount of about 56% (w/w). 5. The implant of claim 1 , wherein said drug is present in an amount of about 54% (w/w). 6. The implant of claim 1 , wherein said drug is present in an amount of about 52% (w/w). 7. The implant of claim 1 , wherein said drug is present in an amount of about 50% (w/w). 8. The implant of claim 1 , wherein said drug is present in an amount of about 40% (w/w). 9. The implant of claim 1 , wherein said drug is present in an amount of about 30% (w/w). 10. The implant of claim 1 , wherein said drug is present in an amount of about 28% (w/w). 11. The implant of claim 1 , wherein said drug is present in an amount of about 20% (w/w). 12. The implant of claim 1 , wherein said drug is present in an amount of about 15% (w/w). 13. The implant of claim 1 , wherein said drug is present in an amount of about 10% (w/w). 14. The implant of claim 1 , wherein said polymer is a biodegradable polymer. 15. The implant of claim 1 , wherein said polymer is PLGA polymer. 16. The implant of claim 1 , wherein said PLA:PGA molar ratio is 75:25. 17. The implant of claim 1 , wherein said PLA:PGA molar ratio is 80:20. 18. The implant of claim 1 , wherein said PLA:PGA molar ratio is 85:15. 19. The implant of claim 1 , wherein said polymer is PLA. 20. The implant of claim 1 , wherein said polymer is present in an amount of about 40-90% (w/w). 21. The implant of claim 20 , wherein said polymer is present in an amount of about 40-70% (w/w). 22. The implant of claim 20 , wherein said polymer is present in an amount of about 70-90% (w/w). 23. The implant of claim 1 , wherein said polymer is present in an amount of about 30% (w/w). 24. The implant of claim 1 , wherein said polymer is present in an amount of about 35% (w/w). 25. The implant of claim 1 , wherein said polymer is present in an amount of about 40% (w/w). 26. The implant of claim 1 , wherein said polymer is present in an amount of about 50% (w/w). 27. The implant of claim 1 , wherein said implant exhibits an inherent viscosity ranging from about 0.2 dl/g to about 0.9 dl/g. 28. The implant of claim 1 , wherein said implant is a subcutaneous implant. 29. The implant of claim 1 , wherein said implant extends the release duration of said therapeutic drug upon subcutaneous administration in said subject maintaining a therapeutic level of the therapeutic drug for a period of 1 to 6 months.
