The present disclosure relates to self-supporting films for delivery of a therapeutic agent containing at least one hydrophobic polymer and at least one therapeutic agent. Methods of forming the self-supporting films are also disclosed.

What is claimed is: 1. A self-supporting multilayer implantable film comprising: a first continuous film layer comprising at least one hydrophobic polymer, a second discontinuous film layer including a plurality of discrete sections in spaced relation relative to each other and disposed on the first continuous film layer, the second discontinuous film layer comprising at least one water-soluble therapeutic agent, and a third continuous film layer comprising at least one hydrophobic polymer and disposed over the second discontinuous film layer, wherein the second discontinuous film layer is disposed between the first and third continuous layers and the self-supporting multilayer film has a tensile strength ranging from about 500 kPa to about 100 Mpa and a therapeutic payload from about 800 μg/cm 2 to about 50 mg/cm 2 . 2. The self-supporting multilayer film of claim 1 , wherein the at least one hydrophobic polymer is selected from the group consisting of aliphatic polyesters, polyhydroxyalkanoates, polyorthoesters, polyurethanes, polyanhydrides, polymer drugs and combinations thereof. 3. The self-supporting multilayer film of claim 2 , wherein the aliphatic polyester is selected from the group consisting of lactide; glycolide; trimethylene carbonate; Δ-valerolactone; β-butyrolactone; γ-butyrolactone; ε-decalactone; hydroxybutyrate; hydroxyvalerate; 1,4-dioxepan-2-one; 1,5-dioxepan-2-one; 6,6-dimethyl-1,4-dioxan-2-one; 2,5-diketomorpholine; pivalolactone; α, α diethylpropiolactone; ethylene carbonate; ethylene oxalate; 3-methyl-1,4-di oxane-2,5-di one; 3,3-di ethyl-1,4-di oxan-2,5-di one; 6,8-dioxabicycloctane-7-one; and homopolymers and copolymers and combinations thereof. 4. The self-supporting multilayer film of claim 1 , wherein the hydrophobic polymer of the first continuous film layer comprises a copolymer of about 10 weight % glycolide and about 90 weight % ε-caprolactone. 5. The self-supporting multilayer film of claim 4 , wherein the hydrophobic polymer of the third film layer comprises a copolymer of about 10 weight % glycolide and about 90 weight % ε-caprolactone. 6. The self-supporting multilayer film of claim 5 , wherein the second discontinuous layer further comprises at least one hydrophobic polymer comprising a copolymer of about 10 weight % glycolide and about 90 weight % ε-caprolactone. 7. The self-supporting multilayer film of claim 1 , wherein the second discontinuous film layer further comprises at least one hydrophobic polymer selected from the group consisting of aliphatic polyesters, polyhydroxyalkanoates, polyorthoesters, polyurethanes, polyanhydrides, polymer drugs and combinations thereof. 8. The self-supporting multilayer film of claim 1 , wherein the hydrophobic polymer of the third film layer comprises a copolymer of about 10 weight % glycolide and about 90 weight % ε-caprolactone. 9. The self-supporting multilayer film of claim 1 , wherein the multilayer film is formed by spray coating. 10. The self-supporting multilayer film of claim 9 , wherein the spray coating comprises ultrasonic spray coating. 11. The self-supporting multilayer film of claim 1 , wherein the at least one water-soluble therapeutic agent is selected from the group consisting of antimicrobial agents, anesthetics, analgesics, anticancer agents, angiogenic agents, antiseptics, antibiotics, fibrotic agents, antimitotics, chelating agents, peptides, proteins, DNA, RNA, nucleotides, liposomes, blood products, hormones, water-soluble silver salts, growth factors, antibodies, interleukins, cytokines, and combinations thereof. 12. The self-supporting multilayer film of claim 11 , wherein the at least one water-soluble therapeutic agent is selected from the group consisting of bupivacaine, fluorouracil, cisplatin, methotrexate, capsaicin, and combinations thereof. 13. The self-supporting multilayer film of claim 11 , wherein the at least one water-soluble therapeutic agent comprises bupivacaine hydrochloride. 14. The self-supporting multilayer film of claim 1 , wherein the tensile strength ranges from about 1 MPa to about 50 MPa. 15. The self-supporting multilayer film of claim 1 , further comprising a thickness ranging from about 0.1 μm to about 3000 μm. 16. The self-supporting multilayer film of claim 1 , further comprising a thickness ranging from about 10 μm to about 500 μm. 17. The self-supporting film of claim 1 , wherein a first section of the plurality of discrete sections includes a first water-soluble therapeutic agent and a second section of the plurality of discrete sections includes a second water-soluble therapeutic agent that is different from the first water-soluble therapeutic agent. 18. The self-supporting film of claim 17 , wherein the first section includes a first hydrophobic polymer and the second section includes a second hydrophobic polymer that is different from the first hydrophobic polymer. 19. The self-supporting multilayer film of claim 1 , wherein a first surface of the first continuous layer is joined to a first surface of the second discontinuous layer along an entire length of the first surface of the second discontinuous layer and a first surface of the third continuous layer is joined to a second surface of the second discontinuous layer along an entire length of the second surface of the second continuous layer. 20. The self-supporting multilayer film of claim 1 , wherein the second discontinuous film layer is disposed entirely between the first and third continuous layers.
