A novel constrained peptide epitope derived from Aβ, related antibody compositions and methods of use. An isolated antibody that specifically binds to a cyclic peptide comprising the conformational epitope corresponding to a solvent-exposed, antibody accessible knuckle region of oligomeric Aβ is described. An antigenic peptide comprising an epitope having a constrained cyclic configuration, corresponding to a solvent-exposed, antibody accessible knuckle region of oligomeric Aβ is also described. Methods of treating, preventing, and diagnosing Alzheimer's disease are also described.

What is claimed is: 1. An antigenic peptide comprising a conformational epitope having a constrained cyclic configuration, wherein the conformational epitope has an amino acid sequence of at least SNK corresponding to a solvent-exposed, antibody accessible knuckle region of oligomeric Aβ, and wherein the K (Lysine) is solvent-exposed. 2. The antigenic peptide of claim 1 , wherein the conformational epitope has an amino acid sequence corresponding to SEQ ID NO: 1. 3. The antigenic peptide of claim 1 or claim 2 , wherein the solvent-exposed, antibody accessible knuckle region of oligomeric Aβ corresponds to residues 25 to 29 of oligomeric Aβ (1-40) or residues 25 to 29 of oligomeric Aβ (1-42). 4. The antigenic peptide of claim 1 , wherein the antigenic peptide has an amino acid sequence of SEQ ID NO: 8. 5. An immunogenic composition comprising the antigenic peptide of claim 1 and a pharmaceutically acceptable adjuvant. 6. A method of treating or delaying the onset of Alzheimer's Disease in a patient in need of said treatment comprising administering said immunogenic composition of claim 5 . 7. An immunogenic composition comprising the antigenic peptide of claim 3 and a pharmaceutically acceptable adjuvant. 8. A method of treating or delaying the onset of Alzheimer's Disease in a patient in need of said treatment comprising administering said immunogenic composition of claim 7 . 9. The immunogenic composition of claim 5 , wherein the conformational epitope of the antigenic peptide has an amino acid sequence corresponding to SEQ ID NO: 1. 10. A method of treating or delaying the onset of Alzheimer's Disease in a patient in need of said treatment comprising administering said immunogenic composition of claim 9 . 11. The immunogenic composition of claim 5 , wherein the antigenic peptide has an amino acid sequence corresponding to SEQ ID NO: 8. 12. A method of treating or delaying the onset of Alzheimer's Disease in a patient in need of said treatment comprising administering said immunogenic composition of claim 11 .
