Disclosed herein are isolated human monoclonal antibodies, and functional fragments thereof, that specifically bind HMW-MAA. Nucleic acids encoding these antibodies, expression vectors including these nucleic acid molecules, and isolated host cells that express the nucleic acid molecules are also disclosed. The antibodies can be used to detect HMW-MAA in a sample. Methods of diagnosing cancer, or confirming a diagnosis of cancer, are disclosed herein that utilize these antibodies. Methods of treating a subject with cancer are also disclosed.

The invention claimed is: 1. An isolated nucleic acid molecule encoding a HMW-MAA-specific monoclonal antibody or antigen-binding fragment thereof, wherein the heavy chain variable region of the antibody or antigen-binding fragment thereof comprises amino acids 27-38, 56-55 and 105-115 of SEQ ID NO: 5, and the light chain variable region of the antibody or antigen-binding fragment thereof comprises amino acids 27-38, 56-65 and 105-110 of SEQ ID NO: 6. 2. The isolated nucleic acid molecule of claim 1 , wherein the heavy chain variable region of the antibody or antigen-binding fragment thereof comprises SEQ ID NO: 5. 3. The isolated nucleic acid molecule of claim 1 , wherein the light chain variable region of the antibody or antigen-binding fragment thereof comprises SEQ ID NO: 6. 4. The isolated nucleic acid molecule of claim 1 , wherein the heavy chain variable region of the antibody or antigen-binding fragment thereof comprises SEQ ID NO: 5, and the light chain variable region of the antibody or antigen-binding fragment thereof comprises SEQ ID NO: 6. 5. The isolated nucleic acid molecule of claim 1 operably linked to a promoter. 6. An expression vector comprising the isolated nucleic acid molecule of claim 1 . 7. An isolated host cell transformed with the expression vector of claim 6 . 8. An isolated host cell transformed with the nucleic acid molecule of claim 1 .
