Abstract

Official abstract text for this publication.

Compositions and methods are provided for treating diseases associated with CD100, including certain autoimmune diseases, inflammatory diseases, and cancers. In particular, anti-CD100 monoclonal antibodies have been developed to neutralize CD100.

First claim

What is claimed is: 1. An isolated antibody or antigen-binding fragment thereof comprising a heavy chain variable region (VH) and a light chain variable region (VL), wherein the VH comprises the amino acid sequence SEQ ID NO: 9, and wherein the antibody or fragment thereof can specifically bind to CD100. 2. The antibody or fragment thereof of claim 1 , wherein the VL comprises an amino acid sequence identical to SEQ ID NO: 17, except for five or fewer conservative amino acid substitutions. 3. The antibody or fragment thereof of claim 2 , wherein the VL comprises an amino acid sequence identical to SEQ ID NO: 17, except for two or fewer conservative amino acid substitutions. 4. The antibody fragment of claim 1 , which is an Fab fragment, an Fab′ fragment, an F(ab′)2 fragment, an Fd fragment, a single-chain Fv fragment (scFv), or a disulfide-linked Fv fragment (sdFv). 5. The antibody or fragment thereof of claim 1 , which comprises a light chain constant region selected from the group consisting of a human kappa constant region and a human lambda constant region. 6. The antibody or fragment thereof of claim 1 , which comprises a heavy chain constant region or fragment thereof. 7. The antibody or fragment thereof of claim 6 , wherein said heavy chain constant region or fragment thereof is human IgG1, IgG2, IgG3, IgG4, IgM, IgA1, IgA2, IgE or IgD. 8. The antibody or fragment thereof of claim 1 , which binds to human and murine CD100. 9. The antibody or fragment thereof of claim 1 , which inhibits CD100 from binding to a CD100 receptor. 10. The antibody or fragment thereof of claim 9 , wherein said CD100 receptor is Plexin-B1. 11. The antibody or fragment thereof of claim 1 , which is conjugated to a cytotoxic agent, a therapeutic agent, a cytostatic agent, a biological toxin, a prodrug, a peptide, a protein, an enzyme, a virus, a lipid, a biological response modifier, a pharmaceutical agent, a lymphokine, a heterologous antibody or fragment thereof, a detectable label, polyethylene glycol (PEG), or any combination thereof. 12. The antibody or fragment thereof of claim 11 , wherein the cytotoxic agent is a radionuclide, a biotoxin, an enzymatically active toxin, or any combination thereof. 13. The antibody or fragment thereof of claim 11 , wherein said detectable label is an enzyme, a fluorescent label, a chemiluminescent label, a bioluminescent label, a radioactive label, or a combination thereof. 14. A composition comprising the antibody or fragment thereof of claim 1 , and a carrier. 15. An isolated antibody or antigen-binding fragment thereof comprising a heavy chain variable region (VH) and a light chain variable region (VL), wherein the VL comprises the amino acid sequence SEQ ID NO: 17, and wherein the antibody or fragment thereof can specifically bind to CD100. 16. The antibody or fragment thereof of claim 15 , wherein the VH comprises an amino acid sequence identical to SEQ ID NO: 9, except for five or fewer conservative amino acid substitutions. 17. The antibody or fragment thereof of claim 16 , wherein the VH comprises an amino acid sequence identical to SEQ ID NO: 9, except for two or fewer conservative amino acid substitutions. 18. The antibody fragment of claim 15 , which is an Fab fragment, an Fab′ fragment, an F(ab′)2 fragment, an Fd fragment, a single-chain Fv fragment (scFv), or a disulfide-linked Fv fragment (sdFv). 19. The antibody or fragment thereof of claim 15 , which comprises a light chain constant region selected from the group consisting of a human kappa constant region and a human lambda constant region. 20. The antibody or fragment thereof of claim 15 , which comprises a heavy chain constant region or fragment thereof. 21. The antibody or fragment thereof of claim 20 , wherein said heavy chain constant region or fragment thereof is human IgG1, IgG2, IgG3, IgG4, IgM, IgA1, IgA2, IgE or IgD. 22. The antibody or fragment thereof of claim 15 , which binds to human and murine CD100. 23. The antibody or fragment thereof of claim 15 , which inhibits CD100 from binding to a CD100 receptor. 24. The antibody or fragment thereof of claim 23 , wherein said CD100 receptor is Plexin-B1. 25. The antibody or fragment thereof of claim 15 , which is conjugated to a cytotoxic agent, a therapeutic agent, a cytostatic agent, a biological toxin, a prodrug, a peptide, a protein, an enzyme, a virus, a lipid, a biological response modifier, a pharmaceutical agent, a lymphokine, a heterologous antibody or fragment thereof, a detectable label, polyethylene glycol (PEG), or any combination thereof. 26. The antibody or fragment thereof of claim 25 , wherein the cytotoxic agent is a radionuclide, a biotoxin, an enzymatically active toxin, or any combination thereof. 27. The antibody or fragment thereof of claim 25 , wherein said detectable label is an enzyme, a fluorescent label, a chemiluminescent label, a bioluminescent label, a radioactive label, or a combination thereof. 28. A composition comprising the antibody or fragment thereof of claim 15 , and a carrier.
