This disclosure concerns plants, plant materials, and the oil characteristics thereof. In embodiments, compositions and methods for determining the total oil content of plant materials are provided. Such methods and compositions may in some embodiments allow for the characterization and screening of oil traits and sub-traits that are difficult or impossible to distinguish by conventional techniques.

What is claimed is: 1. A method for identifying a plant comprising at least one oil trait or sub-trait, the method comprising: (a) providing a dried and ground sample of a plant material; (b) extracting oil from the dried and ground sample of the plant material; (c) admixing the extracted oil with a base, so as to saponify esters in the extracted oil and thereby produce glycerol; (d) reacting the glycerol with an enzymatic glycerol detection reagent to stoichiometrically convert the glycerol to a colored reaction product; (e) colorimetric analyzing the color reaction product to determine an amount of the glycerol in the sample and consequently an amount of oil in the plant material; performing the steps (a) to (e) where the dried and ground sample is taken from a plant having a trait of high-oil content, to determine an amount of oil in the plant having a trait of high-oil content; performing the steps (a) to (e) where the dried and ground sample is taken from a plant having a trait of normal-oil content, to determine an amount of oil in the plant having a trait of normal-oil content; performing the steps (a) to (e) where the dried and ground sample is taken from a plant of interest, to determine an amount of oil in the plant of interest; and comparing the amount of oil in the plant of interest with the amount of oil in the plant having a trait of high-oil content and the amount of oil in the plant having a trait of normal-oil content to determine whether the plant of interest comprises the trait of high-oil content or normal-oil content. 2. The method according to claim 1 , wherein the plant material is seed and the sample is an individual seed. 3. The method according to claim 1 , wherein the plant material is seed and the sample is an individual seed embryo. 4. The method according to claim 1 , wherein the plant material is obtained from a plant selected from a group consisting of corn, soya, canola, sunflower, and cotton. 5. The method according to claim 1 , wherein the plant material is selected from a group consisting of vegetative plant material, seed, leaves, and roots. 6. The method according to claim 5 , wherein the plant material is seed. 7. The method according to claim 6 , wherein the plant material is an embryo. 8. The method according to claim 7 , wherein the sample is a single plant seed embryo. 9. The method according to claim 1 , wherein the sample is too small to provide accurate determination of the total oil content in the plant material by a method selected from the group consisting of gravimetric extraction of oil by an organic solvent and pulsed nuclear magnetic resonance (NMR) spectroscopy. 10. The method according to claim 1 , wherein the sample is lyophilized. 11. The method according to claim 1 , wherein extracting oil from the dried and ground sample of the plant material comprises extracting the oil from the sample using heptane as a solvent. 12. The method according to claim 1 , wherein the base is an alkaline metal salt. 13. The method according to claim 12 , wherein the base is KOH or NaOH. 14. The method according to claim 1 , wherein colorimetric analyzing the color reaction product to determine the amount of glycerol in the sample comprises comparing data obtained with the sample and data obtained with an oil standard having a known oil content. 15. The method according to claim 14 , wherein the oil standard having a known oil content is a refined, bleached, and deodorized oil standard.
