Abstract

Official abstract text for this publication.

Antibodies and molecules derived therefrom that bind to novel STEAP-1 protein, and variants thereof, are described wherein STEAP-1 exhibits tissue specific expression in normal adult tissue, and is aberrantly expressed in the cancers listed in Table I. Consequently, STEAP-1 provides a diagnostic, prognostic, prophylactic and/or therapeutic target for cancer. The STEAP-1 gene or fragment thereof, or its encoded protein, or variants thereof, or a fragment thereof, can be used to elicit a humoral or cellular immune response; antibodies or T cells reactive with STEAP-1 can be used in active or passive immunization.

First claim

The invention claimed is: 1. An isolated polynucleotide encoding a monoclonal antibody that binds to the STEAP-1 protein of SEQ ID NO:3, wherein the antibody comprises all heavy and light chain complementarity determining regions (CDRs) from the antibody designated X120.545.1.1 (ATCC Accession number PTA-5803). 2. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody is the monoclonal antibody designated X120.545.1.1 (ATCC Accession number PTA-5803). 3. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody is a humanized antibody. 4. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody is an antigen binding antibody fragment. 5. The isolated polynucleotide of claim 4 , wherein the antibody fragment is an Fab, F(ab′)2, Fv or Sfv fragment. 6. An isolated polynucleotide encoding a monoclonal antibody which is a humanized form of the monoclonal antibody designated X120.545.1.1 (ATCC Accession number PTA-5803), wherein the monoclonal antibody specifically binds to the STEAP-1 protein of SEQ ID NO:3. 7. An isolated polynucleotide encoding a recombinant protein comprising the antigen binding fragment of a monoclonal antibody that binds to the STEAP-1 protein of SEQ ID NO:3, wherein the antibody comprises all heavy and light chain complementarity determining regions (CDRs) from the antibody designated X120.545.1.1 (ATCC Accession number PTA-5803). 8. An isolated polynucleotide encoding a recombinant protein comprising the antigen binding fragment of a monoclonal antibody which is a humanized form of the monoclonal antibody designated X120.545.1.1 (ATCC Accession number PTA-5803), wherein the monoclonal antibody specifically binds to the STEAP-1 protein of SEQ ID NO:3. 9. An isolated vector comprising the polynucleotide of claim 1 . 10. An isolated vector comprising the polynucleotide of claim 6 . 11. An isolated vector comprising the polynucleotide of claim 7 . 12. An isolated vector comprising the polynucleotide of claim 8 . 13. An isolated host cell comprising the vector of claim 9 . 14. An isolated host cell comprising the vector of claim 10 . 15. An isolated host cell comprising the vector of claim 11 . 16. An isolated host cell comprising the vector of claim 12 .
