The present disclosure is directed to progastrin monoclonal antibodies, fragments thereof, compositions comprising progastrin monoclonal antibodies, and methods of making and using progastrin monoclonal antibodies and compositions thereof. The present disclosure is directed to methods of treating colorectal cancer with progastrin monoclonal antibodies and compositions comprising progastrin monoclonal antibodies or fragments thereof. The present disclosure is further directed to methods comprising detection of progastrin, including methods of diagnosing colorectal cancer and methods of monitoring efficacy of anti-cancer therapy in subjects suffering from colorectal cancer.

What is claimed is: 1. A monoclonal antibody that specifically binds a human progastrin (hPG) polypeptide having an amino acid sequence of SEQ ID NO:20, but does not detectably bind to an amidated gastrin 17 consisting of SEQ ID NO:103, a glycine-extended gastrin 17 consisting of SEQ ID NO: 104, or C-terminal Flanking Peptide (CTFP) consisting of SEQ ID NO:105, wherein said monoclonal antibody is a C-terminal anti-hPG monoclonal antibody that specifically binds a C-terminal region of hPG, wherein: (a) the antibody is obtainable using an immunogen comprising a peptide antigen having an amino acid sequence of SEQ ID NO: 27; and (b) the antibody comprises V H CDRs comprising amino acid sequences of V H CDR 1.8 (SEQ ID NO:37), V H CDR 2.8 (SEQ ID NO:41), V H CDR 3.8 (SEQ ID NO:45) and V L CDRs comprising amino acid sequences of V L CDR 1.8 (SEQ ID NO:49), V L CDR 2.8 (SEQ ID NO:52), and V L CDR 3.8 (SEQ ID NO:55). 2. A monoclonal antibody that specifically binds a human progastrin (hPG) polypeptide having an amino acid sequence of SEQ ID NO:20, but does not detectably bind to an amidated gastrin 17 consisting of SEQ ID NO: 103, a glycine-extended gastrin 17 consisting of SEQ ID NO: 104, or C-terminal Flanking Peptide (CTFP) consisting of SEQ ID NO:105, wherein said monoclonal antibody is a C-terminal anti-hPG monoclonal antibody that specifically binds a C-terminal region of hPG, wherein: (a) the antibody is obtainable using an immunogen comprising a peptide antigen having an amino acid sequence of SEQ ID NO: 27; and (b) the antibody comprises V L and V H CDRs having the amino acid sequences of the V L and V H CDRs of an antibody comprising a V H of SEQ ID NO:59 and a V L of SEQ ID NO:63. 3. A monoclonal antibody that specifically binds a human progastrin (hPG) polypeptide having an amino acid sequence of SEQ ID NO:20, but does not detectably bind to an amidated gastrin 17 consisting of SEQ ID NO:103, a glycine-extended gastrin 17 consisting of SEQ ID NO: 104, or C-terminal Flanking Peptide (CTFP) consisting of SEQ ID NO: 105, wherein said monoclonal antibody is a C-terminal anti-hPG monoclonal antibody that specifically binds a C-terminal region of hPG, wherein: (a) the antibody is obtainable using an immunogen comprising a peptide antigen having an amino acid sequence of SEQ ID NO: 27; and (b) the antibody comprises V H and V L chains having sequences selected from one of the following groups of V H and V L sequences: (i) hV H 8a (SEQ ID NO:75) and hV L 8a (SEQ ID NO:76); (ii) hV H 8b (SEQ ID NO:77) and hV L 8b (SEQ ID NO:78); and (iii) hV H 8c (SEQ ID NO:79) and hV L 8c (SEQ ID NO:76). 4. A monoclonal antibody that specifically binds a human progastrin polypeptide and comprises a V H chain comprising CDRs having the amino acid sequence of V H CDR 1.8 (SEQ ID NO:37), V H CDR 2.8 (SEQ ID NO:41), V H CDR 3.8 (SEQ ID NO:45), and a V L chain comprising CDRs having the amino acid sequence of V L CDR 1.8 (SEQ ID NO:49), V L CDR 2.8 (SEQ ID NO:52), and V L CDR 3.8 (SEQ ID NO:55). 5. The monoclonal antibody of claim 4 , which is humanized. 6. The monoclonal antibody of claim 5 , which comprises V H and V L chains having amino acid sequences of (i) hV H 8a (SEQ ID NO:75) and hV L 8a (SEQ ID NO:76); (ii) hV H 8b (SEQ ID NO:77) and hV L 8b (SEQ ID NO:78); or hV H 8c (SEQ ID NO:79) and hV L 8c (SEQ ID NO:76). 7. The monoclonal antibody of claim 4 , which reduces the binding of a reference antibody to hPG by at least 40% when said monoclonal antibody and the reference antibody are each used at a concentration of 10 μg/ml. 8. The monoclonal antibody of claim 4 , which neutralizes hPG activity in an in vitro assay carried out with LS174T cells, wherein said in vitro assay comprises treating LS174T cells with 5 μg/ml of said monoclonal antibody every 12 hours for 48 hours and determining whether the number of live LS174T cells is reduced by at least 40% as compared to LS174T cells treated with a control antibody. 9. A composition comprising a monoclonal antibody according to claim 4 and an excipient, carrier, and/or diluent. 10. The composition of claim 9 which is formulated for pharmaceutical use. 11. A polynucleotide encoding a variable light chain for a monoclonal antibody according to claim 4 . 12. A polynucleotide encoding a variable heavy chain for an a monoclonal antibody according to claim 4 . 13. An expression vector comprising a polynucleotide encoding a variable light chain for a monoclonal antibody according to claim 4 . 14. An expression vector comprising a polynucleotide encoding a variable heavy chain for a monoclonal antibody according to claim 4 . 15. A cell line transformed with pairs of polynucleotides suitable for expressing a monoclonal antibody according to claim 4 . 16. A cell line capable of secreting an anti-hPG monoclonal antibody according to claim 4 . 17. A method of obtaining a monoclonal antibody according to claim 4 comprising: (a) culturing a cell line of claim 15 under suitable conditions; and (b) recovering said monoclonal antibody from the culture medium or cells of the cell line. 18. A kit useful for detecting human progastrin polypeptide (hPG), comprising a monoclonal antibody according to claim 4 and an antibody that specifically binds an N-terminal region of hPG. 19. The kit of claim 18 , wherein the antibody that specifically binds an N-terminal region of hPG is a polyclonal antibody. 20. A cell line which recombinantly expresses an anti-human progastrin antibody comprising a V H of SEQ ID NO:59 and a V L of SEQ ID NO:63.
