The present relation relates to recombinant vesicular stomatitis virus for use as prophylactic and therapeutic vaccines for infectious diseases of AIDS. The present invention encompasses the preparation and purification of immunogenic compositions which are formulated into the vaccines of the present invention.

What is claimed is: 1. A recombinant vesicular stomatitis virus (VSV) vector encoding and expressing a membrane-anchored HIV Env immunogen and a natural VSV attachment protein (G) (EnvG) chimera, wherein the genes in the VSV vector are arranged sequentially in the order 3′-N-P-M-EnvG-L-G-5′. 2. The vector of claim 1 , wherein the G gene is transferred to a 5′-terminal position in the negative-sense RNA genome of the recombinant VSV vector. 3. The vector of claim 1 , wherein the vector comprises a sequence selected from the group consisting of SEQ ID NO: 5 and SEQ ID NO: 6. 4. The vector of claim 1 , wherein the vector expresses a sequence selected from the group consisting of SEQ ID NO: 1, SEQ ID NO: 3 and SEQ ID NO: 4. 5. The vector of claim 1 , wherein the EnvG chimera comprises a truncated cytoplasmic tail. 6. The vector of claim 2 , wherein the EnvG chimera comprises a truncated cytoplasmic tail. 7. A method of producing an immune response or eliciting an immune response comprising administering to a mammal the vector of claim 1 .
