A mutant MMLV reverse transcriptase that may have an improvement in one or more properties is provided. For example, the present reverse transcriptase is believed to be more efficient relative to other commercially available MMLV reverse transcriptase variants, particularly for templates with a higher GC content.

What is claimed is: 1. A polypeptide comprising at least 300 contiguous amino acids of SEQ ID NO:1. 2. The polypeptide of claim 1 , wherein the polypeptide comprises amino acid residues 24-335 of SEQ ID NO:1. 3. The polypeptide of claim 1 , wherein the polypeptide comprises a truncated N-terminus relative to SEQ ID NO:1. 4. The polypeptide of claim 1 , wherein the polypeptide comprises the entire contiguous sequence of SEQ ID NO:1. 5. The polypeptide of claim 1 , wherein the polypeptide comprises an amino acid sequence that is at least 90% identical to at least 300 contiguous amino acids of SEQ ID NO:2 that is C-terminal to the at least 300 contiguous amino acids of SEQ ID NO:1. 6. The polypeptide of claim 5 , wherein the polypeptide comprises an exogenous sequence-specific DNA binding domain. 7. The polypeptide of claim 5 , wherein the polypeptide comprises a purification tag. 8. The polypeptide of claim 5 , wherein the polypeptide has reverse transcriptase activity. 9. The polypeptide of claim 8 , wherein the polypeptide does not have RNAseH activity. 10. The polypeptide of claim 8 , wherein the polypeptide has RNAseH activity.
