Systems, including methods, apparatus, and algorithms, for spectrally imaging a two-dimensional array of samples.
Opening claim text (preview).
What we claim: 1. A system for sample analysis, comprising: an examination area including a two-dimensional array of examination sites; a light detector for imaging; an optical relay structure that spectrally disperses light received from the examination sites into a corresponding two-dimensional array of spectra directed concurrently onto the light detector; and a thermal control system operatively coupled to the examination area for regulating a temperature thereof; wherein each spectra is disposed parallel to a single spectral axis. 2. The system of claim 1 , wherein the optical element comprises a light guide that operates by total internal reflection. 3. The system of claim 1 , wherein optical relay structure comprises an excitation relay structure, the excitation relay structure comprising the optical element comprises a light guide that operates by total internal reflection. 4. The system of claim 1 , wherein optical relay structure comprises an emission relay structure, the emission relay structure comprising the optical element comprises a light guide that operates by total internal reflection. 5. The system of claim 1 , further comprising a controller operatively coupled to the light detector and configured to process image data collected by the light detector to correct for optical distortion in the image data. 6. The system of claim 1 , wherein, during use, the light detector collects image data of the two-dimensional array of spectra. 7. The system of claim 1 , wherein the optical relay structure comprises at least one of a prism or grating that spectrally disperses light received from the examination sites. 8. A method of sample analysis, comprising: providing an examination area including a two-dimensional array of samples of nucleic acids; providing a thermal control system operatively coupled to the examination area for regulating a temperature thereof; using an excitation relay structure, illuminating each of the samples with light from a light source concurrently such that the samples emit light; and using an emission relay structure, spectrally dispersing light received from the examination sites into a corresponding two-dimensional array of spectra directed concurrently onto a light detector; wherein each spectra is disposed parallel to a single spectral axis. 9. The method of claim 8 , wherein the optical element comprises a light guide that operates by total internal reflection. 10. The method of claim 8 , wherein the excitation relay structure comprises the optical element and illuminating each sample comprises illuminating each sample via the optical element. 11. The method of claim 8 , wherein the emission relay structure comprises the optical element and the optical element is configured to relay the light from the examination sites to the light detector. 12. The method of claim 8 , further comprising collecting image data of the two-dimensional array of spectra. 13. The method of claim 8 , wherein spectrally dispersing light comprises at least one of a prism or grating that spectrally disperses light received from the examination sites.
