The system includes a simple system for CD4 and CD8 counting in point-of-care HIV staging within resource poor countries. Unlike previous approaches, no sample preparation is required with the sample added directly to a chip containing dried reagents by capillary flow. A large area image cytometer consisting of an LED module is used to excite the fluorochromes PerCP and APC labeled targets and a monochrome CCD camera with a combination of two macro lenses captures images of 40 mm 2 of blood (approximately 1 micro liter). CD4 and CD8-T-lymphocyte counts correlate well with those obtained by flow cytometry. The cytometer system described in the present invention provides an affordable and easy-to-use technique for use in remote locations.

What is claimed is: 1. A sample preparation method for combining reagents within a blood sample in order to detect cells using image cytometry, comprising: a. adding at least one reagent comprising a fluorochrome conjugated antibody to a microfluidic chamber having a height between approximately 5 μm and 150 μm wherein the reagent is contained in a dried hydrogel to provide a controlled release of the reagent within the chamber upon contact with the sample; b. filling the chamber with a blood sample wherein the reagent is released from the dried hydrogel into the sample between a time required to fill the chamber and a time required to stain the cells; c. distributing the hydrogel containing the reagent within the chamber such that during the time required to stain the cells the released reagent is uniformly distributed within an imaging area of the chamber; and d. releasing the reagent from the hydrogel within the chamber in order to detect blood cells in the blood sample using image cytometry. 2. The method of claim 1 where the reagent-containing hydrogel releases the reagents by dissolving in the blood sample. 3. The method of claim 1 wherein the reagent molecules are within a reagent layer approximately 150 nm thick. 4. The method of claim 1 where blood fills the chamber between approximately 0.5 s and 5 s. 5. The method of claim 1 where the hydrogel is gelatin. 6. The method of claim 1 where the reagent is released in between 10 s 10 minutes. 7. The method of claim 1 where the reagent is released from the hydrogel with a change in temperature. 8. The method of claim 1 where the blood contains cells which are stained by the reagent. 9. The method of claim 1 where the fluorochrome is Peridinin-Chlorophyll-Protein. 10. The method of claim 1 where the fluorochrome is allophycocyanin. 11. The method of claim 1 where the chamber has a maximum height of approximately 100 μm. 12. A sample preparation method for combining reagents within a blood sample in order to detect cells using image cytometry, comprising; a. adding at least a Peridinin-Chlorophyll-Protein conjugated antibody reagent and an allophycocyanin conjugated antibody reagent to a microfluldic chamber having a height smaller than the diffusion length of the reagent within a 30 min incubation time through the blood sample and larger than approximately 5 μm wherein the reagents are contained in a dried gelatin to provide a controlled release of the reagent within the chamber upon contact with the blood sample; b. filling the chamber with a blood sample wherein at least 50% of the reagents in the dried gelatin are released between a time required to fill the chamber and a time required to stain the cells; c. distributing the gelatin with bound reagents within the chamber such that during the time required to stain the cells the released reagents are uniformly distributed within an imaging area of the chamber; and d. releasing the reagents from the gelatin within the chamber in order to detect blood cells in the, blood sample using image cytometry.
