Disclosed are immunoconjugates having in particular specificity for CD138 expressed on target cells and which display homogenous targeting. The immunoconjugates may be sterially hindered and/or contain a cleavable linker.

First claim

What is claimed is: 1. An immunoconjugate comprising: (a) a targeting antibody, and (b) an effector molecule, wherein the effector is linked to the targeting antibody via a SPDB (N-succinimidyl-4-(2-pyridyldithio)butyrate linker and wherein said immunoconjugate targets CD138 expressing cells, wherein the targeting antibody comprises: heavy chain variable region CDR3 comprising amino acid residues 99 to 111 of SEQ ID NO: 1, and light chain variable region CDR3 comprising amino acid residues 89 to 97 of SEQ ID NO: 2, respectively, and heavy chain variable region CDR1 and CDR2 comprising amino acid residues 31 to 35 and 51 to 68 of SEQ ID NO: 1, and light chain variable region CDR1 and CDR2 comprising amino acid residues 24 to 34 and 50 to 56 of SEQ ID NO: 2, respectively. 2. The immunoconjugate of claim 1 , wherein a constant region of said heavy chain is an IgG4 isotype constant region. 3. The immunoconjugate of claim 1 , comprising said heavy chain variable region CDR1 and CDR2 and said light chain variable region CDR1 and CDR2, respectively. 4. The immunoconjuage according to claim 1 , wherein the immunoconjugate has a K D value of less than 2.6. 5. The immunoconjugate of claim 1 , wherein the effector molecule is at least one maytansinoid, taxane or a CC1065, or an analog thereof. 6. The immunconjugate of claim 5 , wherein the effector molecule is at least one maytansinoid. 7. The immunconjugate of claim 6 , wherein the at least one maytansinoid is DM1, DM3, or DM4. 8. The immunconjugate claim 7 wherein said effector molecule is DM4. 9. An immunoconjugate comprising: an effector molecule, a targeting antibody targeting CD138 comprising an isolated polypeptide comprising amino acid residues 31 to 35, 51 to 68 and 99 to 111 of SEQ ID NO: 1 and amino acid residues 24 to 34, 50 to 56 and 89 to 97 of SEQ ID NO: 2, wherein the effector molecule is linked to the targeting antibody via a SPDB (N-succinimidyl-4-(2-pyridyldithio)butyrate) linker. 10. The immunoconjugate of claim 9 , wherein the targeting antibody comprises SEQ ID NO:1 and SEQ ID NO.2 and the effector molecule is a maytansinoid. 11. The immunoconjugate of claim 10 , wherein the effector molecule is DM1 or DM4. 12. The immunoconjugate of claim 1 , wherein the targeting antibody comprises a sequence having at least 95% sequence identity with SEQ ID: NO:1 or at least 90% sequence identity with SEQ ID NO:2 and further comprises all of said heavy and light chain variable regions. 13. The immunoconjugate of claim 1 , wherein the targeting antibody comprises SEQ ID: NO:1 and/or SEQ ID NO:2. 14. The immunconjugate of claim 1 , wherein the engineered targeting antibody comprises sequences having at least 95% sequence identity with SEQ ID: NO:1 and at least 90% sequence identity with SEQ ID NO:2 and further comprises all of said heavy and light chain variable regions. 15. The immunoconjugate of claim 1 , wherein the immunconjugate comprises SEQ ID: NO:1 and SEQ ID NO:2. 16. The immunoconjugate of claim 11 , wherein the effector molecule is DM4. 17. The immunoconjugate of claim 4 , wherein the antibody is a chimerized antibody. 18. An immunoconjugate comprising: DM4 as an effector molecule, a targeting antibody targeting CD138 comprising an immunoglobin heavy and light chain, the immunoglobulin heavy chain being encoded by SEQ ID NO:1 and the immunoglobulin light chain being encoded by SEQ ID NO. 2, wherein the constant region of said immunoglobulin heavy chain is an IgG4 isotype constant region, and wherein the DM4 effector molecule is linked to said targeting antibody via a SPDB (N-succinimidyl-4-(2-pyridyldithio)butyrate) linker.
