The present invention provides a method of optical electrophysiological probing, including: providing a fluorescing chemical probe; contacting a thick portion of tissue with the fluorescing chemical probe to create a thick portion of treated tissue; applying a first range of wavelengths of electromagnetic radiation to the treated portion of tissue; and detecting a plurality of depth-specific emission wavelengths emitted from the thick portion of treated tissue.

We claim: 1. An optically mapping composition comprising a voltage-sensitive dye of the formula: wherein R 1 is a first hydrocarbon group that is a butyl group; R 2 is a second hydrocarbon group that is a butyl group; and R 3 is a butyl sulfonate polar group. 2. The composition of claim 1 , further comprising a delivery agent that is at least one of a physiologically acceptable adjuvant, solvent, substrate, or diluent. 3. The composition of claim 2 , wherein the delivery agent is selected from the group consisting of: Pluronic F-127, Pluronic L64, cyclodextrin, gamma cyclodextrin, dimethylsulfoxide, ethanol, and combinations thereof.
