This invention provides an antibody targeting a cancer antigenic protein specifically expressed on the surface of cancer cells and use thereof as a therapeutic and/or preventive agent for cancer. More specifically, the present invention provides an antibody, or a fragment thereof which has immunological reactivity with a partial CAPRIN-1 polypeptide consisting of the amino acid sequence shown by SEQ ID NO: 5 or an amino acid sequence having 80% or higher sequence identity to the amino acid sequence, and a pharmaceutical composition for treatment and/or prevention of cancer comprising the same as an active ingredient.

First claim

The invention claimed is: 1. An antibody or a fragment thereof which has immunological reactivity with a partial CAPRIN-1 polypeptide consisting of the amino acid sequence shown by SEQ ID NO: 5. 2. The antibody or the fragment thereof according to claim 1 , wherein the antibody or the fragment thereof has cytotoxic activity against a cancer cell expressing a CAPRIN-1 protein. 3. The antibody or the fragment thereof according to claim 1 , wherein the antibody is a monoclonal antibody or a polyclonal antibody. 4. The antibody or the fragment thereof according to claim 1 , wherein the antibody is a human antibody, a humanized antibody, a chimeric antibody, a single-chain antibody, or a multispecific antibody. 5. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 6, 7, and 8 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 10, 11, and 12 and has immunological reactivity with the CAPRIN-1 protein. 6. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 16, 17, and 18 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 20, 21, and 22 and has immunological reactivity with the CAPRIN-1 protein. 7. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 6, 7, and 8 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 50, 51, and 52 and has immunological reactivity with the CAPRIN-1 protein. 8. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 55, 56, and 57 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 59, 60, and 61 and has immunological reactivity with the CAPRIN-1 protein. 9. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 55, 56, and 57 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 59, 64, and 61 and has immunological reactivity with the CAPRIN-1 protein. 10. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 66, 67, and 68 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 70, 71, and 72 and has immunological reactivity with the CAPRIN-1 protein. 11. The antibody or the fragment thereof according to claim 1 , which comprises a heavy chain variable region comprising complementarity determining regions of SEQ ID NOs: 74, 75, and 76 and a light chain variable region comprising complementarity determining regions of SEQ ID NOs: 78, 79, and 80 and has immunological reactivity with the CAPRIN-1 protein. 12. The antibody or the fragment thereof according to claim 1 , wherein the antibody or the fragment thereof is conjugated with an antitumor agent. 13. A pharmaceutical composition for treatment of cancer, comprising the antibody or the fragment thereof according to claim 1 as an active ingredient, wherein the cancer is a CAPRIN-1 expressing cancer. 14. The pharmaceutical composition according to claim 13 , wherein the cancer is breast cancer, kidney cancer, pancreatic cancer, colorectal cancer, lung cancer, brain tumor, gastric cancer, uterine cervix cancer, ovary cancer, prostate cancer, urinary bladder cancer, esophageal cancer, leukemia, lymphoma, fibrosarcoma, mastocytoma, or melanoma. 15. A pharmaceutical combination for treatment of cancer, comprising a pharmaceutical composition according to claim 13 and a pharmaceutical composition comprising an antitumor agent. 16. A DNA encoding the antibody or the fragment thereof according to claim 1 . 17. A method for treating cancer, comprising administering the antibody or the fragment thereof according to claim 1 , wherein the cancer is a CAPRIN-1 expressing cancer. 18. The antibody or the fragment thereof according to claim 2 , wherein the antibody is a monoclonal antibody or a polyclonal antibody. 19. The antibody or the fragment thereof according to claim 2 , wherein the antibody is a human antibody, a humanized antibody, a chimeric antibody, a single-chain antibody, or a multispecific antibody. 20. The antibody or the fragment thereof according to claim 3 , wherein the antibody is a human antibody, a humanized antibody, a chimeric antibody, a single-chain antibody, or a multispecific antibody.
