The present invention relates to isolated polypeptides having cellobiohydrolase activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

First claim

The invention claimed is: 1. A nucleic acid construct or expression vector comprising a polynucleotide encoding a polypeptide having cellobiohydrolase activity and one or more heterologous control sequences that direct production of the polypeptide in an expression host, wherein the polynucleotide is operably linked to the one or more heterologous control sequences that direct production of the polypeptide in an expression host, and wherein the polypeptide having cellobiohydrolase activity comprises an amino acid sequence that has at least 92% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 2. An isolated recombinant host cell comprising the nucleic acid construct or expression vector of claim 1 . 3. A method of producing a polypeptide having cellobiohydrolase activity, comprising: (a) cultivating the recombinant host cell of claim 2 under conditions conducive for production of the polypeptide; and (b) recovering the polypeptide. 4. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity further comprises a cellulose binding domain. 5. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity is a variant of the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2, wherein the variant is modified by a substitution, deletion, and/or insertion of one or more amino acids in the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2, and wherein the polypeptide having cellobiohydrolase activity comprises an amino acid sequence that has at least 92% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 6. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 95% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 7. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 8. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises or consists of the amino acid sequence of SEQ ID NO: 2. 9. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 97% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 10. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 98% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 11. The nucleic acid construct or expression vector of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 99% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 12. An isolated recombinant host cell transformed with a polynucleotide encoding a polypeptide having cellobiohydrolase activity, wherein the polypeptide having cellobiohydrolase activity has at least 92% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2, and wherein the polynucleotide is heterologous to the recombinant host cell. 13. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity has at least 95% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 14. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity has at least 97% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 15. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity has at least 98% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 16. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity has at least 99% sequence identity to the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 17. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity comprises the amino acid sequence of amino acids 19 to 455 of SEQ ID NO: 2. 18. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity comprises or consists of the amino acid sequence of SEQ ID NO: 2. 19. The isolated recombinant host cell of claim 12 , wherein the polypeptide having cellobiohydrolase activity further comprises a cellulose binding domain. 20.
