Nucleic acid molecules encoding chimeric cellulase polypeptides that exhibit improved cellulase activities are disclosed herein. The chimeric cellulase polypeptides encoded by these nucleic acids and methods to produce the cellulases are also described, along with methods of using chimeric cellulases for the conversion of cellulose to sugars such as glucose.

An isolated nucleic acid molecule encoding a chimeric CbhA polypeptide comprising domains from Clostridium thermocellum cellobiohydrolase A (CbhA) and Caldicellulosiruptor bescii cellobiohydrolase A (CelA) polypeptides, wherein the chimeric CbhA polypeptide has an amino acid sequence at least 95% identical to “SEQ ID NO:6,” and wherein the chimeric CbhA polypeptide has a cellulase activity at least 1.5-fold greater than the wild-type CbhA polypeptide.…
