The present invention relates to compositions and methods for characterizing, diagnosing, and treating cancer. In particular the invention provides the means and methods for the diagnosis, characterization, prognosis and treatment of cancer and specifically targeting cancer stem cells. The present invention provides an antibody that specifically binds to a non-ligand binding membrane proximal region of the extracellular domain of a human Notch receptor and inhibits tumor growth. The present invention further provides a method of treating cancer, the method comprising administering a therapeutically effective amount of an antibody that specifically binds to a non-ligand binding membrane proximal region of the extracellular domain of a human Notch receptor protein and inhibits tumor growth.

What is claimed is: 1. An isolated polynucleotide encoding an antibody that specifically binds human NOTCH1, wherein the antibody comprises: (a) a heavy chain CDR1 comprising RGYWIE (SEQ ID NO:15); a heavy chain CDR2 comprising QILPGTGRTNYNEKFKG (SEQ ID NO:16); and a heavy chain CDR3 comprising FDGNYGYYAMDY (SEQ ID NO:17); and (b) a light chain CDR1 comprising RSSTGAVTTSNYAN (SEQ ID NO:18); a light chain CDR2 comprising GTNNRAP (SEQ ID NO:19); and a light chain CDR3 comprising ALWYSNHWVFGGGTKL (SEQ ID NO:20). 2. The isolated polynucleotide of claim 1 , wherein the antibody is a monoclonal antibody, a chimeric antibody, a humanized antibody, a human antibody, or an antibody fragment. 3. A vector comprising the polynucleotide of claim 1 . 4. An isolated cell comprising the vector of claim 3 . 5. An isolated cell comprising the polynucleotide of claim 1 . 6. The isolated polynucleotide of claim 1 , wherein the antibody comprises: (a) a heavy chain variable region having at least 90% sequence identity to SEQ ID NO:14 or SEQ ID NO:24; and (b) a light chain variable region having at least 90% sequence identity to SEQ ID NO:8, SEQ ID NO:28, or SEQ ID NO:32. 7. The polynucleotide of claim 6 , wherein the antibody comprises: (a) a heavy chain variable region having at least about 90% sequence identity to SEQ ID NO:24; and (b) a light chain variable region having at least about 90% sequence identity to SEQ ID NO:28 or SEQ ID NO:32. 8. The polynucleotide of claim 7 , wherein the antibody comprises: (a) a heavy chain variable region comprising SEQ ID NO:24; and (b) a light chain variable region comprising SEQ ID NO:28. 9. The polynucleotide of claim 7 , wherein the antibody comprises: (a) a heavy chain variable region comprising SEQ ID NO:24; and (b) a light chain variable region comprising SEQ ID NO:32. 10. The polynucleotide of claim 6 , wherein the antibody comprises: (a) a heavy chain variable region having at least about 90% sequence identity to SEQ ID NO:14; and (b) a light chain variable region having at least about 90% sequence identity to SEQ ID NO:8. 11. The polynucleotide of claim 10 , wherein the antibody comprises: (a) a heavy chain variable region comprising SEQ ID NO:14; and (b) a light chain variable region comprising SEQ ID NO:8. 12. The isolated polynucleotide of claim 6 , wherein the antibody is a monoclonal antibody, a chimeric antibody, a humanized antibody, a human antibody, or an antibody fragment. 13. A vector comprising the polynucleotide of claim 6 . 14. An isolated cell comprising the vector of claim 13 . 15. An isolated cell comprising the polynucleotide of claim 6 . 16. An isolated polynucleotide encoding a polypeptide comprising the sequence of: (a) SEQ ID NO:22, SEQ ID NO:23, SEQ ID NO:24, SEQ ID NO:10, SEQ ID NO:12, or SEQ ID NO:14; and (b) SEQ ID NO:26, SEQ ID NO:27, SEQ ID NO:28, SEQ ID NO:30, SEQ ID NO:31, SEQ ID NO:32, SEQ ID NO:4, SEQ ID NO:6, or SEQ ID NO:8. 17. The isolated polynucleotide of claim 16 , wherein the polynucleotide encodes a polypeptide comprising the sequence of SEQ ID NO:24 and SEQ ID NO:28. 18. The isolated polynucleotide of claim 16 , wherein the polynucleotide encodes a polypeptide comprising the sequence of SEQ ID NO:22 and SEQ ID NO:26. 19. An isolated polynucleotide comprising the sequence of: (a) SEQ ID NO:21, SEQ ID NO:9, SEQ ID NO:11, or SEQ ID NO:13; and (b) SEQ ID NO:25, SEQ ID NO:29, SEQ ID NO:3, SEQ ID NO:5, or SEQ ID NO:7. 20. The isolated polynucleotide of claim 19 , comprising the sequence of SEQ ID NO:21 and SEQ ID NO:25. 21. An isolated polynucleotide deposited with ATCC as PTA-9549. 22. An isolated polynucleotide encoding the hybridoma deposited with ATCC as PTA-9405.
