A production method for stage 5 neurons with mature dendrites includes culturing stage 3 neurons with immature dendrites in a culture medium containing B-27 Plus.
Opening claim text (preview).
What is claimed is: 1 . A production method for stage 5 neurons with mature dendrites comprising: culturing stage 3 neurons with immature dendrites in a culture medium containing B-27 Plus. 2 . The production method according to claim 1 , wherein the stage 5 neurons have dendritic spines. 3 . The production method according to claim 1 , wherein a postsynaptic structure including drebrin and PSD95 is formed in the stage 5 neurons. 4 . The production method according to claim 1 , wherein the stage 3 neurons express a neuronal marker including MAP2 and neurofilament heavy chain (NF-H1) and are polarized. 5 . The production method according to claim 1 , wherein the stage 3 neurons are obtained by differentiation induction of induced pluripotent stem cells (iPSCs). 6 . The production method according to claim 5 , wherein the stage 3 neurons are induced to differentiate by induction of expression of a transcription factor in the iPSCs. 7 . Stage 5 neurons with mature dendrites that are isolated and cultured in a container. 8 . A screening method for a therapeutic drug for a neurological disease, comprising: culturing stage 3 neurons that are induced to differentiate from neurological disease patient-derived iPSCs in a culture medium containing B-27 Plus in the presence of a test substance to obtain stage 5 neurons; and assessing a phenotype of the stage 5 neurons, wherein a case in which the phenotype of the stage 5 neurons in the presence of the test substance exhibits a phenotype more similar to a wild-type as compared with a phenotype of the stage 5 neurons in the absence of the test substance indicates that the test substance is a candidate of the therapeutic drug for the neurological disease. 9 . A screening method for a therapeutic drug for a neurological disease, comprising: culturing stage 5 neurons that are induced to differentiate from neurological disease patient-derived iPSCs for a predetermined period of time in the presence of a test substance; and assessing a phenotype of the stage 5 neurons, wherein a case in which the phenotype of the stage 5 neurons in the presence of the test substance exhibits a phenotype more similar to a wild-type as compared with a phenotype of the stage 5 neurons in the absence of the test substance indicates that the test substance is a candidate of the therapeutic drug for the neurological disease.
