Described are single domain antibodies with a specificity for BACE1. More specifically, described are single variable-domain antibodies derived from camelids that bind to BACE1 and are capable of inhibiting the activity of BACE1. The antibodies can be used for research and medical applications. Specific applications include the use of BACE1-specific antibodies for the treatment of Alzheimer's disease.

A single domain antibody, devoid of a light chain, wherein the single domain antibody specifically binds beta secretase (BACE1) and inhibits cleavage of beta amyloid precursor protein (APP) in an endosomal compartment of a cell as determined in a cellular assay when the single-domain antibody is provided to the extracellular medium of the cell in the cellular assay. 11 . The single domain antibody of claim 10 , wherein the single domain antibody is derived from an antibody raised in a Camelidae species. 12 . A medicament comprising: the single domain antibody of claim 10 ; and at least one pharmaceutically acceptable carrier. 13 . The single-domain antibody of claim 10 , wherein the single-domain antibody is a VHH antibody. 14 . The single-domain antibody of claim 10 , wherein the single-domain antibody comprises four framework regions (FRs) and three complementary determining regions (CDRs). 15 . The single-domain antibody of claim 14 , wherein the four FRs and three CDRs are in sequence: FR-1-CDR1-FR2-CDR2-FR3-CDR3-FR4. 16 . The single-domain antibody of claim 10 , wherein the antibody is bivalent. 17 . The single-domain antibody of claim 10 , wherein the single-domain antibody is bi-specific.
