Provided herein are compositions and methods for modulation of immune response via PYRIN domain-only proteins POP1 and/or POP3. In particular, POP1 and/or POP3 are inhibited to enhance an immune response (e.g., to treat or prevent infection), or POP1 and/or POP3 are administered or activated to reduce an immune response (e.g., to treat or prevent autoimmune or inflammatory disease).

1 . A method of treating or preventing a disease or condition associated with inflammation comprising administering a composition that increases the activity of POP1, POP2, and/or POP3. 2 . The method of claim 1 , wherein the composition comprises a POP1, POP2, and/or POP3 polypeptide, peptide, or peptidomimitic. 3 . The method of claim 2 , wherein the polypeptide or peptide comprises 100% sequence identity with all or a portion of POP1, POP2, or POP3. 4 . The method of claim 2 , wherein the polypeptide or peptide comprises less than 100% sequence identity, but greater than 70% sequence identity with all or a portion of POP1, POP2, or POP3. 5 . The method of claim 1 , wherein the composition comprises an agent that enhances the inflammasome inhibitory activity of POP1, POP2, and/or POP3. 6 . The method of claim 5 , wherein the composition is a small molecule, peptide, polypeptide, protein, or antibody. 7 . The method of claim 1 , wherein the disease or condition is an autoimmune disease. 8 . The method of claim 1 , wherein the disease or condition is an inflammatory disease. 9 . The method of claim 1 , wherein the composition is co-administered with one or more additional treatments for the disease or condition 10 . A method of treating or preventing infection comprising administering a composition that inhibits the activity of POP1, POP2, and/or POP3. 11 . The method of claim 10 , wherein the composition is a small molecule, peptide, polypeptide, protein, or antibody. 12 . The POP1, POP2, or POP3 protein, polypeptide, or peptide that comprises less than 100% sequence identity, but greater than 70% sequence identity with all or a portion of POP1, POP2, or POP3. 13 . A POP1, POP2, or POP3 protein, polypeptide, or peptide for use as a medicament. 14 . The POP1, POP2, or POP3 protein, polypeptide, or peptide of claim 13 , comprising less than 100% sequence identity, but greater than 70% sequence identity with all or a portion of POP1 or POP3. 15 . The POP1, POP2, or POP3 protein, polypeptide, or peptide of claim 13 for use in the treatment or prevention of an autoimmune or inflammatory disease or condition. 16 . A POP1, POP2, or POP3 protein, polypeptide, or peptide for use in the treatment or prevention of an autoimmune or inflammatory disease or condition. 17 . A POP1, POP2, or POP3 inhibitor. 13 .
