This disclosure is directed to an apparatus, system and method for retrieving a target material from a sample. A density-altering agent may be added to a vessel that contains the sample to change the density of a non-target material.

First claim

I/We claim: 1 . A method for collecting a target material from a sample, the method comprising the steps of: adding a density-altering agent to the sample to change the density of at least a first non-target material relative to the density of the target material; inserting a collector into an open end of a primary vessel that contains the suspension, the collector including a cavity in a first end, an opening in a second end, and a cannula to extend between the opening and the cavity; inserting a processing vessel into the cavity, the cannula to extend into the processing vessel; adding a collection fluid to the processing vessel, the collection fluid having a density greater than the density of the target material; and centrifuging the primary vessel, the collector, the collection fluid, and the processing vessel, wherein during centrifuging, the collection fluid flows into the primary vessel via the collector to displace the target material from the primary vessel into the processing vessel through the collector via the cannula. 2 . The method of claim 1 , further comprising the step of: separating the suspension into suspension fractions prior to adding the density-altering agent to the sample. 3 . The method of claim 2 , further comprising the step of: adding a depletion agent to the sample to change the density of at least a second non-target material relative to the density of the target material, wherein the depletion agent is added prior to the separating step. 4 . The system of claim 3 , wherein the depletion agent makes the density of platelets greater than or equal to red blood cells, and wherein the target material is at least one circulating tumor cell or fetal material. 5 . The method of claim 2 , further comprising the step of: removing at least a portion of a non-target material from the primary vessel after the separating step and before inserting the collector into the primary vessel. 6 . The method of claim 5 , further comprising the step of: adding a clearing fluid to the primary vessel after the removing step, the clearing fluid having a density greater than at least the target material. 7 . The method of claim 6 , further comprising the step of: adding a float to the primary vessel prior to the separating step. 8 . The method of claim 7 , further comprising the step of: forming a first seal between the float and the primary vessel by clamping after the separating step, the removing step, or adding the clearing fluid. 9 . The method of claim 8 , further comprising the step of: centrifuging the primary vessel after forming the first seal. 10 . The method of claim 9 , further comprising the step of: forming a second seal between the float and the primary vessel by clamping after the centrifuging step performed after forming the first seal, wherein the first seal is formed between a lower end of the main body of the float and the inner wall of the primary vessel, and wherein the second seal is formed between an upper end of the main body of the float and the inner wall of the primary vessel. 11 . The method of claim 1 , further comprising the step of: forming a seal between a second end of the collector and an inner wall of the primary vessel to maintain a fluid-tight sealing engagement between the collector and inner wall of the primary vessel. 12 . The method of claim 1 , wherein the collection fluid is selected from the group consisting of: a solution of colloidal silica particles coated with polyvinylpyrrolidone, a polysaccharide solution, iodixanol, an organic solvent, a liquid wax, an oil, a gas, olive oil, mineral oil, silicone oil, immersion oil, mineral oil, paraffin oil, silicon oil, fluorosilicone, perfluorodecalin, perfluoroperhydrophenanthrene, perfluorooctylbromide, organic solvents, 1,4-Dioxane, acetonitrile, ethyl acetate, tert-butanol, cyclohexanone, methylene chloride, tert-Amyl alcohol, tert-Butyl methyl ether, butyl acetate, hexanol, nitrobenzene, toluene, octanol, octane, propylene carbonate, tetramethylene sulfones, ionic liquids, a polymer-based solution, a surfactant, a perfluoroketone, perfluorocyclopentanone, perfluorocyclohexanone, a fluorinated ketone, a hydrofluoroether, a hydrofluorocarbon, a perfluorocarbon, a perfluoropolyether, silicon, a silicon-based liquid, phenylmethyl siloxane, and combinations thereof. 13 . The method of claim 1 , wherein the processing vessel includes a plug in a closed end, the cannula to extend through the plug. 14 . The method of claim 13 , wherein the plug is composed of a resealeable material. 15 . The method of claim 1 , further comprising the step of: adding at least one antibody to the sample to bind to at least one non-target material, wherein the at least one antibody is conjugated to a first complementary molecule, wherein the density-altering agent is conjugated to a second complementary molecule, and wherein the first and second complementary molecules bind to one another. 16 . The method of claim 1 , further comprising the step of: adding a plurality of antibodies to the sample to bind to at least one non-target material, wherein each of the plurality of antibodies is conjugated to a first complementary molecule, wherein the density-altering agent is conjugated to a second complementary molecule, and wherein the first and second complementary molecules bind to one another. 17 . The method of claim 16 , wherein the plurality of antibodies includes at least two of anti-CD3, anti-CD14, anti-CD33, anti-CD42b, anti-CD45, anti-CD66b, and anti-CD88. 18 . The method of claim 1 , wherein the density-altering agent is conjugated to at least one antibody to bind directly to at least one non-target material. 19 . The method of claim 1 , further comprising the step of: adding a depletion agent to the sample to change the density of at least a second non-target material relative to the density of the target material. 20 . The method of claim 1 , wherein the density-altering agent is at least one particle composed of silica, glass, a polymer, or a metal.
