The present invention is directed to therapeutic methods using IL-6 antagonists such as antibodies and fragments thereof having binding specificity for IL-6 to prevent or treat thrombosis in diseases associated with abnormal blood coagulation or fibrinolysis. In preferred embodiments these patients will comprise those exhibiting elevated D-dimer or other coagulation cascade related proteins and optionally will further exhibit elevated C reactive protein prior to treatment. The subject therapies also may include the administration of other actives such as chemotherapeutics, anti-coagulants, statins, et al.

A method of treating a patient having a disease or condition associated with hypercoagulation, comprising administering to the patient an IL-6 or anti-IL-6R antagonist antibody or antibody fragment, whereby the patient's coagulation profile is improved or restored to normal, and monitoring the patient to assess coagulation profile. 92 . The method of claim 91 wherein the patient has elevated serum D-dimer levels prior to treatment, and/or elevated C-reactive protein (CRP) levels prior to treatment. 93 . The method of claim 91 wherein said method further comprises the administration of at least one statin or anti-coagulant. 94 . The method of claim 94 wherein the IL-6 antagonist comprises an anti-IL-6 antibody or an antibody fragment. 95 . The method of claim 94 wherein the anti-IL-6 antibody or antibody fragment contains the same CDRs as an anti-IL-6 antibody selected from Ab1, Ab2, Ab3, Ab4, Ab5, Ab6, Ab7, Ab8, Ab9, Ab10, Ab11, Ab12, Ab13, Ab14, Ab15, Ab16, Ab17, Ab18, Ab19, Ab20, Ab21, Ab22, Ab23, Ab24, Ab25, Ab26, Ab27, Ab28, Ab29, Ab30, Ab31, Ab32, Ab33, Ab34, Ab35, or Ab36. 96 . The method of claim 95 wherein the anti-IL-6 antibody or antibody fragment is aglycosylated. 97 . The method of claim 91 wherein the antibody comprises the heavy and light constant regions in SEQ ID NO:588 and SEQ ID NO:586. 98 . The method of claim 91 wherein the anti-IL-6 antibody or antibody fragment contains an Fc region that has been modified to alter effector function, half-life, proteolysis, and/or glycosylation. 99 . The method of claim 91 wherein the anti-IL-6 antibody or antibody fragment is a human, humanized, single chain or chimeric antibody. 100 . The method of claim 91 wherein the patient has been diagnosed with cancer. 101 .
