A combination of a CDK4/6 inhibitor and an mTOR inhibitor for the treatment of cancer.

A combination comprising a first agent that is a cyclin dependent kinase 4 or cyclin dependent kinase 6 (CDK4/6) inhibitor and a second agent that is an mTOR inhibitor, wherein the first agent is 7-Cyclopentyl-2-(5-piperazin-1-yl-pyridin-2-ylamino)-7H-pyrrolo[2,3-d]pyrimidine-6-carboxylic acid dimethylamide or a pharmaceutically acceptable salt thereof. 54 . The combination of claim 53 , wherein the second agent is selected from the group consisting of rapamycin (AY-22989), everolimus, CCI-779, AP-23573, MK-8669, AZD-8055, Ku-0063794, OSI-027, WYE-125132. 55 . The combination of claim 54 , wherein the second agent is everolimus. 56 . The combination of claim 53 , wherein the first agent and the second agent are in a combined dosage form. 57 . The combination of claim 53 wherein the first agent and the second agent are in separate dosage forms. 58 . A method of treating cancer comprising administering a first agent that is a cyclin dependent kinase 4 or cyclin dependent kinase 6 (CDK4/6) inhibitor and a second agent that is an mTOR inhibitor wherein the first agent is 7-Cyclopentyl-2-(5-piperazin-1-yl-pyridin-2-ylamino)-7H-pyrrolo[2,3-d]pyrimidine-6-carboxylic acid dimethylamide or a pharmaceutically acceptable salt thereof. 59 . The method of claim 58 wherein the cancer is dependent on the CDK4, CDK6 or mTOR pathway. 60 . The method of claim 59 wherein the cancer is a solid tumor cancer. 61 . The method of claim 58 , wherein the cancer is pancreatic cancer, breast cancer, mantle cell lyomphoma, non-small cell lung cancer, melanoma, colon cancer, esophageal cancer, liposarcoma, multiple myeloma, T-cell leukemia, renal cell carcinoma, gastric cancer, renal cell carcinoma, glioblastoma, hepatocellular carcinoma, gastric cancer, lung cancer or colon cancer. 62 . The method of claim 61 , wherein the cancer is pancreatic cancer, breast cancer, or mantle cell lyomphoma. 63 . The method of claim 58 , wherein the cancer is a lymphoma. 64 . The method of claim 58 wherein the first agent and the second agent are administered in a combined dosage form. 65 . The method of claim 58 wherein the first agent and the second agent are administered in separate dosage forms. 66 . The method of claim 58 wherein the second agent is everolimus.
