Disclosed are methods, compounds, and compositions useful for increasing autophagy and promoting longevity. The methods, compounds, and compositions relate to urolithins and urolithin precursors and use thereof. Certain urolithins are represented by Formula I, while certain urolithin precursors are represented by Formula IV. The urolithin may be urolithin A, urolithin B, urolithin C, or urolithin D. The urolithin precursor may be ellagic acid or an ellagitannin. The methods include in vivo, ex vivo, and in vitro uses of the compounds and compositions.

We claim: 1 . A composition, comprising urolithin A or a pharmaceutically acceptable salt thereof; and a second ingredient selected from the group consisting of resveratrol and coenzyme Q10; wherein the coenzyme Q10 is ubiquinol or ubiquinone. 2 . The composition of claim 1 , wherein the second ingredient is resveratrol. 3 . The composition of claim 1 , wherein the second ingredient is coenzyme Q10; and the coenzyme Q10 is ubiquinol. 4 . The composition of claim 1 , wherein the second ingredient is coenzyme Q10; and the coenzyme Q10 is ubiquinone. 5 . The composition of claim 1 , further comprising a pharmaceutically acceptable carrier. 6 . A method of improving or increasing mitochondrial function, comprising administering to a vertebrate in need thereof an effective amount of the composition of claim 1 . 7 . The method of claim 6 , wherein the mitochondrial function is mitophagy. 8 . The composition of claim 2 , further comprising a pharmaceutically acceptable carrier. 9 . The composition of claim 3 , further comprising a pharmaceutically acceptable carrier. 10 . The composition of claim 4 , further comprising a pharmaceutically acceptable carrier. 11 . A method of improving or increasing mitochondrial function, comprising administering to a vertebrate in need thereof an effective amount of the composition of claim 2 . 12 . The method of claim 11 , wherein the mitochondrial function is mitophagy. 13 . A method of improving or increasing mitochondrial function, comprising administering to a vertebrate in need thereof an effective amount of the composition of claim 3 . 14 . The method of claim 13 , wherein the mitochondrial function is mitophagy. 15 . A method of improving or increasing mitochondrial function, comprising administering to a vertebrate in need thereof an effective amount of the composition of claim 4 . 16 . The method of claim 15 , wherein the mitochondrial function is mitophagy. 17 . The composition of claim 2 , wherein the composition is in the form of a dietary supplement or a nutraceutical. 18 .
