The present invention relates to anti-VEGF-A/anti-ANG2 antibodies, e.g. in the form of a bispecific Fab fragment, and methods of using the same.

The invention claimed is: 1 . An isolated single nucleic acid encoding a Fab heavy chain and a Fab light chain of a Fab fragment that binds to human vascular endothelial growth factor A (VEGF-A) and to human angiopoietin-2 (ANG2), wherein the Fab heavy chain comprises a VH amino acid sequence of SEQ ID NO: 19 and the Fab light chain comprises a VL amino acid sequence of SEQ ID NO: 20. 2 . A host cell comprising an expression vector comprising the nucleic acid of claim 1 . 3 . A method of producing a Fab fragment that binds to human VEGF-A and to human ANG2 comprising culturing the host cell of claim 2 so that the Fab fragment is produced. 4 . The method of claim 3 , wherein the host cell is a CHO cell. 5 . An isolated single nucleic acid encoding a Fab heavy chain and a Fab light chain of a Fab fragment that binds to human VEGF-A and to human ANG2, wherein the Fab heavy chain amino acid sequence is SEQ ID NO: 24 and the Fab light chain amino acid sequence is SEQ ID NO: 25. 6 . A host cell comprising an expression vector comprising the nucleic acid of claim 5 . 7 . A method of producing an antibody a Fab fragment that binds to human VEGF-A and to human ANG2 comprising culturing the host cell of claim 6 so that the Fab fragment is produced. 8 . The method of claim 7 , wherein the host cell is a CHO cell. 9 . An first isolated nucleic acid encoding a Fab heavy chain of a Fab fragment and a second isolated nucleic acid encoding a Fab light chain of a Fab fragment, wherein the Fab fragment binds to human vascular endothelial growth factor A (VEGF-A) and to human angiopoietin-2 (ANG2), wherein the Fab heavy chain comprises a VH amino acid sequence of SEQ ID NO: 19 and the Fab light chain comprises a VL amino acid sequence of SEQ ID NO: 20. 10 . A host cell comprising an expression vector comprising the first isolated nucleic acid and the second isolated nucleic acid of claim 9 . 11 . A method of producing a Fab fragment that binds to human VEGF-A and to human ANG2 comprising culturing the host cell of claim 10 so that the Fab fragment is produced. 12 . The method of claim 11 , wherein the host cell is a CHO cell. 13 . An first isolated nucleic acid encoding a Fab heavy chain of a Fab fragment and a second isolated nucleic acid encoding a Fab light chain of a Fab fragment, wherein the Fab fragment binds to human vascular endothelial growth factor A (VEGF-A) and to human angiopoietin-2 (ANG2), wherein the Fab heavy chain comprises a VH amino acid sequence of SEQ ID NO: 24 and the Fab light chain comprises a VL amino acid sequence of SEQ ID NO: 25. 14 . A host cell comprising an expression vector comprising the first isolated nucleic acid and the second isolated nucleic acid of claim 13 . 15 . A method of producing a Fab fragment that binds to human VEGF-A and to human ANG2 comprising culturing the host cell of claim 14 so that the Fab fragment is produced. 16 . The method of claim 15 , wherein the host cell is a CHO cell.
