Provided herein are Mycobacterium smegmatis porin nanopores, systems that comprise these nanopores, and methods of using and making these nanopores. Such nanopores may be wild-type MspA porins, mutant MspA porins, wild-type MspA paralog porins, wild-type MspA homolog porins, mutant MspA paralog porins, mutant MspA homolog porins, or single-chain Msp porins. Also provided are bacterial strains capable of inducible Msp porin expression.

First claim

The embodiments of the invention in which an exclusive property or privilege is claimed are defined as follows: 1 . A Mycobacterium smegmatis porin (Msp) comprising: a vestibule and a constriction zone that define a tunnel, and at least a first Mycobacterium smegmatis porin A (MspA) monomer variant of SEQ ID NO: 1 comprising: (i) a mutation at position 93 and a mutation at position 90 or position 91, or both positions 90 and 91, wherein the mutation at position 90 is D90N or D90Q and/or the mutation at position 91 is D91N or D91Q; and (ii) one or more mutations at positions 118, 134, and 139 replacing negatively charged amino acids with positively charged amino acids. 2 . The Msp according to claim 1 , wherein the mutant MspA monomer comprises a mutation at positions 93 and 90. 3 . The Msp according to claim 1 , wherein the mutant MspA monomer comprises a mutation at positions 90, 91, and 93. 4 . The Msp according to claim 1 , wherein the mutant MspA monomer comprises a mutation at positions 90, 91, and 93, wherein the mutation at position 90 is D90N, the mutation at position 91 is D91N, and the mutation at position 93 is D93N. 5 . The Msp according to claim 4 , wherein the mutation at position 118 is D118R. 6 . The Msp according to claim 5 , wherein the mutation at position 139 is E139K or E139R. 7 . The Msp according to claim 6 , wherein the mutation at position 118 is D118R and the mutation at position 139 is E139K. 8 . The Msp according to claim 1 , wherein the mutant MspA monomer further comprises a mutation at position 126, wherein the mutation at position 126 is Q126R. 9 . The Msp according to claim 1 , wherein the mutant MspA monomer further comprises a mutation at position 88. 10 . The Msp according to claim 1 , wherein the mutant MspA monomer comprises a mutation at positions 90, 91, and 93, wherein the mutation at position 90 is D90Q, the mutation at position 91 is D91Q, and the mutation at position 93 is D93N. 11 . The Msp according to claim 10 , wherein the mutation at position 118 is D118R, the mutation at position 134 is D134R, and the mutation at position 139 is E139K. 12 . A Mycobacterium smegmatis porin (Msp) comprising: a vestibule and a constriction zone that define a tunnel, and at least a first Mycobacterium smegmatis porin A (MspA) monomer variant of SEQ ID NO: 1, comprising: a mutation at positions 90, 91, and 93, wherein the mutation at position 90 is D90N, the mutation at position 91 is D91N, and the mutation at position 93 is D93N. 13 . A Mycobacterium smegmatis porin (Msp) comprising: a vestibule and a constriction zone that define a tunnel, and at least a first Mycobacterium smegmatis porin A (MspA) monomer variant of SEQ ID NO: 1, comprising: a mutation at positions 90, 91, and 93, wherein the mutation at position 90 is D90Q, the mutation at position 91 is D91Q, and the mutation at position 93 is D93N; and one or more mutations at positions 118, 134, and 139 replacing negatively charged amino acids with positively charged amino acids. 14 . The Msp according to claim 13 , wherein the mutation at position 118 is D118R, the mutation at position 134 is D134R, and the mutation at position 139 is E139K. 15 . A system comprising a Mycobacterium smegmatis porin (Msp) having a vestibule and a constriction zone that define a tunnel, wherein the tunnel is positioned between a first liquid medium and a second liquid medium, wherein the Msp comprises at least a first MspA monomer variant of SEQ ID NO: 1, comprising a mutation at position 93 and a mutation at position 90, position 91, or both positions 90 and 91, wherein the mutation at position 90 is one of D90N or D90Q and/or the mutation at position 91 is one of D91N or D91Q, wherein at least one liquid medium comprises an analyte, wherein the system is operative to detect a property of the analyte. 16 . A system comprising a Mycobacterium smegmatis porin (Msp) having a vestibule and a constriction zone that define a tunnel, wherein the tunnel is positioned between a first liquid medium and a second liquid medium, wherein the Msp comprises at least a first MspA monomer variant of SEQ ID NO: 1, comprising: (i) a mutation at position 93 and a mutation at position 90, position 91, or both positions 90 and 91, wherein the mutation at position 90 is one of D90N or D90Q and/or the mutation at position 91 is one of D91N or D91Q; and (ii) one or more mutations at positions 118, 134, and 139 replacing negatively charged amino acids with positively charged amino acids compared to a wild-type MspA monomer, wherein at least one liquid medium comprises an analyte, wherein the system is operative to detect a property of the analyte.
