Methods are provided for the production of photoreceptor cells and photoreceptor progenitor cells from pluripotent stem cells. Additionally provided are compositions of photoreceptor cells and photoreceptor progenitor cells, as well as methods for the therapeutic use thereof. Exemplary methods may produce substantially pure cultures of photoreceptor cells and/or photoreceptor progenitor cells.

First claim

The invention claimed is: 1 . A preparation comprising: a plurality of cells, wherein at least 85% of the cells are CHX10 (−) photoreceptor progenitor cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and NRO, and wherein the cells are differentiated in vitro from pluripotent stem cells; and a medium suitable for maintaining the viability of the cells, wherein the CHX10 (−) photoreceptor progenitor cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and/or NRO: secrete one or more neuroprotective factors; have phagocytic activity; or a combination thereof. 2 . The preparation of claim 1 , wherein at least 85% of the cells are CHX10 (−) when assessed by immunostaining. 3 . The preparation of claim 1 , wherein at least 90% of the cells are CHX10 (−) by immunostaining. 4 . The preparation of claim 1 , wherein at least 95% of the cells are CHX10 (−) by immunostaining. 5 . The preparation of claim 1 , wherein the CHX10 (−) photoreceptor progenitor cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and NRO secrete one or more neuroprotective factors. 6 . The preparation of claim 1 , wherein the CHX10 (−) cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and NRO have phagocytic activity. 7 . The preparation of claim 1 , wherein the CHX10 (−) cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and NRO: secrete one or more neuroprotective factors; and have phagocytic activity. 8 . The preparation of claim 1 , wherein the CHX10 (−) cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and/or NRO have the ability to phagocytose isolated photoreceptor outer segments, fluorescent bioparticles, or both photoreceptor outer segments and fluorescent bioparticles. 9 . The preparation of claim 1 , wherein the CHX10 (−) cells that express one or more of the markers Nr2e3, Trβ2, Mash1, RORβ and NRO are unable to differentiate into cone photoreceptor cells. 10 . The preparation of claim 1 , wherein the preparation is a pharmaceutical preparation, and wherein the pharmaceutical preparation comprises a pharmaceutically acceptable carrier. 11 . The preparation of claim 1 , wherein the preparation is a cryopreserved preparation, and wherein the cryopreserved preparation comprises a cryopreservative compatible with the viability of the cells. 12 . The preparation of claim 1 , wherein the cells of the preparation are suspended in the medium suitable for maintaining the viability of the cells. 13 . The preparation of claim 1 , further comprises a matrix or a support, wherein the cells of the preparation are attached to the matrix or the support. 14 . The preparation of claim 1 , wherein less than 1% of the cells of the preparation are human embryonic stem cells, induced pluripotent stem cells, eye field progenitor cells, retinal ganglion cells, mature photoreceptors, and/or amacrine cells. 15 . The preparation of claim 1 , wherein the pluripotent stem cells are ES cells. 16 . The preparation of claim 1 , wherein the pluripotent stem cells are iPS cells. 17 . The preparation of claim 1 , wherein the cells are human cells. 18 . The preparation of claim 1 , wherein the cells of the preparation are HLA-matched or genetically identical relative to each other.
