Applicators for applying an on-skin assembly to skin of a host and methods of their use and/or manufacture are provided. An applicator includes an insertion assembly configured to insert at least a portion of the on-skin assembly into the skin of the host, a housing configured to house the insertion assembly, the housing comprising an aperture through which the on-skin assembly can pass, an actuation member configured to, upon activation, cause the insertion assembly to insert at least the portion of the on-skin assembly into the skin of the host, and a sealing element configured to provide a sterile barrier and a vapor barrier between an internal environment of the housing and an external environment of the housing.

What is claimed is: 1 . An apparatus for inserting a transcutaneous glucose sensor, wherein the transcutaneous glucose sensor comprises an in vivo portion and an ex vivo portion, the apparatus comprising: a housing comprising an aperture, wherein the housing is configured for temporary placement against a skin of a host with the aperture facing the skin of the host; an on-skin glucose sensor assembly contained within the housing, wherein the on-skin glucose sensor assembly comprises the transcutaneous glucose sensor and sensor electronics, wherein the ex vivo portion of the transcutaneous glucose sensor is coupled to the sensor electronics, and wherein the on-skin glucose sensor assembly is configured to pass through the aperture in the housing during deployment; an insertion assembly comprising a needle having a C-shaped cross section contained within the housing, wherein the in vivo portion of the transcutaneous glucose sensor is positioned inside the needle, wherein the insertion assembly is configured to insert the in vivo portion of the transcutaneous glucose sensor into the skin of the host using the needle, and wherein the needle and the housing are configured to be removed from the host after insertion with the in vivo portion of the transcutaneous glucose sensor transcutaneously implanted in the host; a cap configured to threadably couple to the housing such that the cap is configured to be removed by twisting the cap with respect to the housing; and an elastomeric support member in contact with an inner surface of the cap. 2 . The apparatus of claim 1 , wherein the elastomeric support member is configured to maintain the insertion assembly in position during deleterious movement including a drop or vibration. 3 . The apparatus of claim 1 , wherein a tamper indicator is positioned over a threaded coupling between the cap and the housing. 4 . The apparatus of claim 3 , wherein the tamper indicator comprises perforations, scoring, or deformed sections. 5 . The apparatus of claim 1 , wherein the housing comprises at least one ridge. 6 . The apparatus of claim 1 , comprising an actuator configured to cause the insertion assembly to push the on-skin glucose sensor assembly through the aperture and insert the in vivo portion of the transcutaneous glucose sensor into the skin of the host. 7 . The apparatus of claim 1 , wherein removing the cap exposes the needle. 8 . The apparatus of claim 1 , wherein the elastomeric support member is configured to absorb shock during deleterious movement including a drop or vibration. 9 . An apparatus for inserting a transcutaneous glucose sensor, wherein the transcutaneous glucose sensor comprises an in vivo portion and an ex vivo portion, the apparatus comprising: a housing comprising an aperture, wherein the housing is configured for temporary placement against a skin of a host with the aperture facing the skin of the host; an on-skin glucose sensor assembly contained within the housing, wherein the on-skin glucose sensor assembly comprises the transcutaneous glucose sensor and sensor electronics, wherein the ex vivo portion of the transcutaneous glucose sensor is coupled to the sensor electronics, and wherein the on-skin glucose sensor assembly is configured to pass through the aperture in the housing during the inserting; an insertion assembly comprising a needle having a C-shaped cross section contained within the housing, wherein the in vivo portion of the transcutaneous glucose sensor is positioned inside the needle, wherein the insertion assembly is configured to insert the in vivo portion of the transcutaneous glucose sensor into the skin of the host using the needle, and wherein the needle and the housing are configured to be removed from the host after insertion with the in vivo portion of the transcutaneous glucose sensor transcutaneously implanted in the host; a sealing element threadably coupled to the housing, wherein the sealing element is configured to be removed by twisting the sealing element with respect to the housing; and wherein the sealing element comprises an inner chamber, an outer chamber, and an elastomeric support member; wherein the in vivo portion of the transcutaneous glucose sensor is positioned inside the inner chamber; wherein the inner chamber comprises an end opening having an outer boundary edge; wherein an annular portion of a surface of the elastomeric support member is engaged with the outer boundary edge of the end opening of the inner chamber of the sealing element. 10 . The apparatus of claim 9 , wherein the sealing element is configured such that twisting the sealing element with respect to the housing decouples the inner chamber, the outer chamber, and the elastomeric support member from the housing to expose the needle. 11 . The apparatus of claim 9 , wherein at least a portion of the sealing element comprises a threaded cap. 12 . The apparatus of claim 9 , wherein the elastomeric support member is not in contact with an insertion end of the needle. 13 . The apparatus of claim 9 , further comprising an actuator configured to cause the insertion assembly to push the on-skin glucose sensor assembly through the aperture and insert the in vivo portion of the transcutaneous glucose sensor into the skin of the host. 14 . The apparatus of claim 9 , wherein a tamper indicator is positioned over the threaded coupling between the sealing element and the housing. 15 . The apparatus of claim 14 , wherein the tamper indicator comprises perforations, scoring, or deformed sections. 16 . The apparatus of claim 9 , wherein at least a portion of the outer chamber is formed by a threaded cap. 17 . The apparatus of claim 9 , wherein the outer chamber at least partially surrounds the inner chamber. 18 . The apparatus of claim 17 , wherein the elastomeric support member forms a boundary between the inner chamber and the outer chamber. 19 . The apparatus of claim 9 , wherein the elastomeric support member forms a boundary between the inner chamber and the outer chamber. 20 . An apparatus for inserting a transcutaneous glucose sensor, wherein the transcutaneous glucose sensor comprises an in vivo portion and an ex vivo portion, the apparatus comprising: a housing comprising an aperture, wherein the housing is configured for temporary placement against a skin of a host with the aperture facing the skin of the host; an on-skin glucose sensor assembly contained within the housing, wherein the on-skin glucose sensor assembly comprises the transcutaneous glucose sensor and sensor electronics, wherein the ex vivo portion of the transcutaneous glucose sensor is coupled to the sensor electronics, and wherein the on-skin glucose sensor assembly is configured to pass through the aperture in the housing during the inserting; an insertion assembly comprising a needle having a C-shaped cross section contained within the housing, wherein the in vivo portion of the transcutaneous glucose sensor is positioned inside the needle, wherein the insertion assembly is configured to insert the in vivo portion of the transcutaneous glucose sensor into the skin of the host using the needle, and wherein the needle and the housing are configured to be removed from the host after insertion with the in vivo portion of the transcutaneous glucose sensor transcutaneously implanted in the host; a cap threadably coupled to the housing such that the cap is configured to be removed by twisting the cap with respect to the housing; and an elastomer
