A method for penetrating a pierceable cap ( 11 ) on a vessel ( 21 ). The pierceable cap ( 11 ) prevents escape of sample specimens from the vessel ( 21 ) before transfer with a transfer device ( 43 ). The pierceable cap ( 11 ) may fit over a vessel ( 21 ). An access port in the shell of the pierceable cap ( 11 ) may allow passage of a transfer device ( 43 ) through the pierceable cap ( 11 ). At least one frangible layer ( 215, 216 ) may be configured with cross slits ( 506 ) in a particular cross slit geometry. The cross slits ( 506 ) may contain an openable portion ( 644 ) or be covered by a thin membrane ( 645 ). The shell ( 610 ) and frangible layer(s) ( 215, 216 ) may be integrated into a one piece cap ( 601 ), or be separate components ( 634 ). The membrane on which the cross slits 506 are placed can be flat or contoured to guide the transfer device ( 43 ) to the cross slits ( 506 ). The flat surfaces are supported by rib structures ( 620, 621 ) that extend radially inward and downward into the vessel ( 21 ).

The invention claimed is: 1. A method for penetrating a pierceable cap comprising: attaching a shell and an elastomeric seal onto a sample vessel, wherein the shell comprises an access port and a threaded portion, wherein the elastomeric seal comprises a collar configured as an annular ring that is positioned between the access port and the threaded portion wherein the annular ring is held in position and compressed when the shell is attached to the sample vessel, and a support band disposed above or below the collar, wherein the support band has a circumference that is slightly less than an outer circumference of the collar of the elastomeric seal; inserting a sample transfer device through the access port in the shell, wherein the shell is attached to the sample vessel by the threaded portion that extends from the access port of the shell, wherein the threaded portion is wider than the access port; advancing the transfer device towards an openable portion disposed on a surface of the elastomeric seal; puncturing the openable portion with the transfer device to access a sample contained within the sample vessel, wherein the elastomeric seal is configured with a radial portion and at least two ribs extending radially inward and axially downward from the radial portion, wherein downward is toward an interior of the vessel; and removing the transfer device from the sample vessel. 2. The method of claim 1 , wherein the shell is elastomeric. 3. The method of claim 1 , wherein the openable portion is a slitted portion and the transfer device is guided to the slitted portion by the at least two ribs. 4. The method of claim 1 , wherein the inserting and advancing of the transfer device is performed by an automated system. 5. The method of claim 4 , wherein the automated system is a robot. 6. The method of claim 1 , wherein the inserting and advancing of the transfer device is performed manually. 7. The method of claim 1 , wherein the openable portion is selected from the group consisting of a tearable slitted portion or an unjoined slit. 8. The method of claim 1 , wherein the access port includes a first cavity. 9. The method of claim 8 , wherein the shell comprises the threaded portion extending from the access port to a distal end of the shell, the threaded portion being dimensioned to define a second cavity that receives the sample vessel. 10. The method of claim 9 , wherein the elastomeric seal comprises the annular ring positioned in a transition between the first cavity and the second cavity, wherein the radial portion is at least partially interposed between the transition and the sample vessel. 11. The method of claim 1 , wherein the support band is monolithically integrated into the elastomeric seal. 12. The method of claim 1 , wherein the support band is an o-ring. 13. The method of claim 12 , wherein the support band is retained in a groove within an inner surface of the shell.
