The present disclosure provides methods and processes for increasing the efficiency and accuracy of nucleic acid sequencing using techniques such as polymerase chain reaction (PCR). Methods and systems provided herein may facilitate performing reactions such as emulsion PCR (ePCR) on samples comprising nucleic acids and beads. The methods provided herein may provide a higher throughput compared to existing technologies.

What is claimed is: 1. A system, comprising: a thermocycler comprising (i) a plurality of fins configured to receive a cartridge comprising a plurality of containers between at least a subset of said plurality of fins and (ii) a fluid channel in thermal communication with said plurality of fins, wherein a cavity defined by at least two of said plurality of fins receives therein at least two containers of said plurality of containers; said cartridge disposed within said thermocycler; a gap between respective walls of said at least two containers and respective walls of said cavity, wherein said gap is subjected to negative pressure to create a vacuum within said gap; and a pump that is configured to subject fluid to flow through said fluid channel, thereby thermocycling a sample in said plurality of containers. 2. The system of claim 1 , wherein said plurality of fins comprise metal. 3. The system of claim 1 , wherein said plurality of fins comprise aluminum. 4. The system of claim 1 , wherein said system comprises a lid configured to close said thermocycler. 5. The system of claim 4 , wherein said lid is configured to maintain a temperature or temperature threshold. 6. The system of claim 1 , wherein said plurality of containers are a plurality of wells. 7. The system of claim 1 , wherein a container of said plurality of containers is configured to hold at least 1 milliliter (1 mL) of fluid. 8. The system of claim 1 , wherein said cavity is configured to receive therein at least four containers of said plurality of containers. 9. The system of claim 8 , wherein said cavity is configured to receive therein at least eight containers of said plurality of containers. 10. The system of claim 9 , wherein said cavity is configured to receive therein at least twelve containers of said plurality of containers. 11. The system of claim 1 , wherein a plurality of cavities is defined by said plurality of fins, and wherein each cavity of said plurality of cavities is configured to receive at least two containers of said plurality of containers. 12. The system of claim 11 , wherein said plurality of cavities are disposed as an array of substantially parallel lanes of cavities. 13. The system of claim 1 , wherein said cavity comprises one or more features configured to guide individual containers of said at least two containers within said cavity. 14. The system of claim 13 , wherein said one or more features are coupled to one or more walls of said plurality of fins. 15. The system of claim 13 , wherein said one or more features are integrated with one or more walls of said plurality of fins. 16. The system of any claim 1 , wherein said cavity comprises a first substantially planar wall, and wherein a container of said at least two containers comprises a second substantially planar wall configured to interface said first substantially planar wall. 17. The system of claim 1 , wherein said vacuum comprises a pressure of at most about 14.7 pounds per square inch (psi).
