Disclosed herein are inhibitors, such as antibodies, and antigen-binding portions thereof, that selectively bind complexes of LTBP1-TGFβ and/or LTBP3-TGFβ. The application also provides methods of use of these inhibitors for, for example, inhibiting TGFβ activation, and treating subjects suffering from TGFβ-related disorders, such as fibrotic conditions. Methods of selecting a context-dependent or context-independent isoform-specific TGFβ inhibitor for a subject in need thereof are also provided.

What is claimed is: 1. A method of inhibiting transforming growth factor β1 (TGFβ1) signaling in a subject, the method comprising administering to the subject an antibody, or an antigen-binding fragment thereof, that specifically binds a human latent transforming growth factor beta binding protein 1 (LTBP1)-proTGFβ1 complex or a human latent transforming growth factor beta binding protein 3 (LTBP3)-proTGFβ1 complex, comprising a heavy chain variable domain and a light chain variable domain, wherein the heavy chain variable domain comprises: a heavy chain complementary determining region 1 (CDRH1) comprising an amino acid sequence set forth as SEQ ID NO: 166, a heavy chain complementary determining region 2 (CDRH2) comprising an amino acid sequence set forth as SEQ ID NO: 167, and a heavy chain complementary determining region 3 (CDRH3) comprising an amino acid sequence set forth as SEQ ID NO: 168; and wherein the light chain variable domain comprises: a light chain complementary determining region 1 (CDRL1) comprising an amino acid sequence set forth as SEQ ID NO: 169, a light chain complementary determining region 2 (CDRL2) comprising an amino acid sequence set forth as SEQ ID NO: 170, and, a light chain complementary determining region 3 (CDRL3) comprising an amino acid sequence set forth as SEQ ID NO: 171, wherein the TGFβ1 is presented by LTBP1 or LTBP3. 2. The method of claim 1 , wherein the heavy chain variable domain comprises an amino acid sequence set forth as SEQ ID NO: 318 and the light chain variable domain comprises an amino acid sequence set forth as SEQ ID NO: 319. 3. The method of claim 1 , wherein the heavy chain variable domain consists of an amino acid sequence set forth as SEQ ID NO: 318 and the light chain variable domain consists of an amino acid sequence set forth as SEQ ID NO: 319. 4. The method of claim 1 , wherein the subject has a fibrotic condition. 5. The method of claim 4 , wherein the fibrotic condition is an organ fibrosis. 6. The method of claim 5 , wherein the organ fibrosis is selected from the group consisting of kidney fibrosis, liver fibrosis, lung fibrosis, cardiac fibrosis, pancreatic fibrosis, skin fibrosis, muscle fibrosis, and uterine fibroids. 7. The method of claim 4 , wherein the fibrotic condition is a fibrotic disorder comprising chronic inflammation. 8. The method of claim 7 , wherein the fibrotic disorder comprising chronic inflammation is muscular dystrophy, multiple sclerosis, or cystic fibrosis. 9. The method of claim 8 , wherein the muscular dystrophy is Duechenne muscular dystrophy (DMD). 10. The method of claim 4 , wherein the fibrotic condition is Alport syndrome. 11. The method of claim 4 , wherein the fibrotic condition is fibrosis, fibroids, desmoplasia, or amyotrophic lateral sclerosis (ALS). 12. The method of claim 4 , wherein the fibrotic condition is a liver disease. 13. The method of claim 12 , wherein the liver disease is selected from the group consisting of non-alcoholic fatty liver disease (NAFLD), stage 2 and 3 liver fibrosis, stage 4 fibrosis, primary biliary cholangitis (PBC), and primary sclerosing cholangitis (PSC). 14. The method of claim 13 , wherein the NAFLD is non-alcoholic fatty liver (NAFL) or non-alcoholic steatohepatitis (NASH). 15. The method of claim 14 , wherein the NASH is noncirrhotic NASH with liver fibrosis, liver cirrhosis, NASH with compensated cirrhosis, NASH with decompensated cirrhosis, or liver inflammation with fibrosis. 16. The method of claim 12 , further comprising administering a myostatin inhibitor to the subject. 17. The method of claim 4 , wherein the fibrotic condition is a kidney disease. 18. The method of claim 17 , wherein the kidney disease is selected from the group consisting of IgA nephropathy, focal and segmental glomerulosclerosis, crescentic glomerulonephritis, lupus nephritis, and diabetic nephropathy. 19. The method of claim 1 , wherein the administering is subcutaneous administration. 20. The method of claim 1 , wherein the heavy chain variable domain comprises an amino acid sequence having at least 95% identity to SEQ ID NO: 318 and the light chain variable domain comprises an amino acid sequence having at least 95% identity to SEQ ID NO: 319.
