A separation container for extracting a portion of a sample for use or testing and method for preparing samples for downstream use or testing are provided. The separation container may include a body defining an internal chamber. The body may define an opening, and the body may be configured to receive the sample within the internal chamber. The separation container may further include a seal disposed across the opening, such that the seal may be configured to seal the opening of the body, and a plunger movably disposed at least partially inside the internal chamber. The plunger may be configured to be actuated to open the seal and express the portion of the sample.

What is claimed is: 1. An assembly comprising: a separation container comprising: a body, wherein the body defines an internal chamber, wherein the body is configured to receive a sample within the internal chamber, and wherein the body defines an opening at a first end of the body; and a seal attached to the body and disposed across the opening, the seal configured to seal the opening at the first end of the body; and an end cap configured to be removably disposed at the first end of the body, the end cap configured to receive the first end of the body such that the seal is positioned between the body and the end cap, wherein the end cap defines an internal cavity, an end cap opening at a first end of the internal cavity, and a support surface at a second end of the internal cavity, wherein the second end is opposite the first end of the internal cavity, wherein the body defines an axis extending from the first end of the body to a second end of the body, wherein the body defines a body flange comprising a flange surface facing parallel to the axis, wherein the end cap defines an end cap flange within the internal cavity configured to engage the body flange of the body. 2. The assembly of claim 1 , wherein the body further defines an annular surface around the opening at the first end of the body, and wherein the seal is attached to the annular surface. 3. The assembly of claim 2 , wherein the end cap defines a support surface configured to be positioned parallel to the annular surface of the body with the seal being disposed between the support surface and the annular surface, wherein the support surface is configured to contact the seal at least during centrifugation to prevent the seal from detaching from the body during centrifugation. 4. The assembly of claim 2 , wherein the seal is welded to the annular surface. 5. The assembly of claim 1 , wherein the end cap opening is defined by a cylindrical wall. 6. The assembly of claim 1 , wherein the body flange is disposed circumferentially about a tapered portion of the body. 7. The assembly of claim 1 , the body and the end cap are configured to form a snap connection therebetween. 8. The assembly of claim 1 , wherein a distal end of the end cap comprises a recess configured to receive a post therein. 9. The assembly of claim 1 , wherein the end cap further comprises one or more projecting ribs extending into an interior cavity of the end cap. 10. The assembly of claim 1 , wherein the seal comprises a liquid-tight membrane. 11. The assembly of claim 1 , wherein the separation container further comprises a coupling member defining a coupling member opening therethrough, wherein the coupling member is disposed around a portion of the body, and wherein the seal and a second portion of the body extends through the coupling member opening. 12. The assembly of claim 1 , wherein the end cap is a centrifuge adapter configured to engage a centrifuge cup to support the body during centrifugation. 13. The assembly of claim 11 , wherein the coupling member comprises an elastomeric sleeve disposed around the portion of the body. 14. A separation container comprising: a body, wherein the body defines an internal chamber, wherein the body is configured to receive a sample within the internal chamber, and wherein the body defines an opening at a first end of the body; a seal attached to the body and disposed across the opening, the seal configured to seal the opening at the first end of the body; and a coupling member defining a coupling member opening therethrough, wherein the coupling member is disposed around a portion of the body, and wherein the seal and a second portion of the body extends through the coupling member opening, wherein the separation container is configured to receive an end cap at the first end of the body, such that the seal is positioned between the body and the end cap. 15. The separation container of claim 14 , wherein the body further defines an annular surface around the opening at the first end of the body, and wherein the seal is attached to the annular surface. 16. The separation container of claim 15 , wherein the seal is welded to the annular surface. 17. The separation container of claim 14 , wherein the coupling member comprises an elastomeric sleeve disposed around the portion of the body. 18. An assembly comprising: a separation container comprising: a body, wherein the body defines an internal chamber, wherein the body is configured to receive a sample within the internal chamber, and wherein the body defines an opening at a first end of the body; and a seal attached to the body and disposed across the opening, the seal configured to seal the opening at the first end of the body; and an end cap configured to be removably disposed at the first end of the body, the end cap configured to receive the first end of the body such that the seal is positioned between the body and the end cap, wherein the end cap further comprises one or more projecting ribs extending into an interior cavity of the end cap.
