The present invention provides binding agents comprising at least two binding domains, wherein a first binding domain has specificity for CLDN18.2 and a second binding domain has specificity for CD3, and methods of using these binding agents or nucleic acids encoding therefor for treating cancer.

The invention claimed is: 1. A bispecific binding agent comprising a first binding domain that binds to human CLDN18.2, a second binding domain that binds to human CLDN18.2, and a third binding domain that binds to human CD3, wherein the bispecific binding agent comprises four polypeptide chains, wherein (i) the first polypeptide chain comprises the amino acid sequence of SEQ ID NO: 73; (ii) the second polypeptide chain comprises the amino acid sequence of SEQ ID NO: 74; (iii) the third polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78; and (iv) the fourth polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78. 2. The bispecific binding agent of claim 1 , wherein the first polypeptide chain consists of the amino acid sequence of SEQ ID NO: 73. 3. The bispecific binding agent of claim 1 , wherein the second polypeptide chain consists of the amino acid sequence of SEQ ID NO: 74. 4. The bispecific binding agent of claim 1 , wherein the third polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78. 5. The bispecific binding agent of claim 1 , wherein the fourth polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78. 6. A bispecific binding agent comprising a first binding domain that binds to human CLDN18.2, a second binding domain that binds to human CLDN18.2, and a third binding domain that binds to human CD3, wherein the bispecific binding agent comprises four polypeptide chains, wherein the first polypeptide chain consists of the amino acid sequence of SEQ ID NO: 73; wherein the second polypeptide chain consists of the amino acid sequence of SEQ ID NO: 74; wherein the third polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78; and wherein the fourth polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78. 7. One or more nucleic acid molecules encoding the bispecific binding agent of claim 1 . 8. The one or more nucleic acid molecules of claim 7 , wherein the one or more nucleic acid molecules is a set of nucleic acids. 9. A host cell comprising the one or more nucleic acid molecules of claim 7 . 10. A vector comprising the one or more nucleic acid molecules of claim 7 . 11. A host cell comprising the vector of claim 10 . 12. One or more nucleic acid molecules comprising: a) a first nucleic acid sequence encoding a first polypeptide chain comprising the amino acid sequence of SEQ ID NO: 73; b) a second nucleic acid sequence encoding a second polypeptide chain comprising the amino acid sequence of SEQ ID NO: 74; and c) a third nucleic acid sequence encoding a third polypeptide chain comprising the amino acid sequence of SEQ ID NO: 78. 13. A host cell comprising the one or more nucleic acid molecules of claim 12 . 14. A set of vectors comprising: i) a first vector comprising a nucleic acid encoding a first polypeptide chain comprising the amino acid sequence of SEQ ID NO: 73; ii) a second vector comprising a nucleic acid encoding a second polypeptide chain comprising the amino acid sequence of SEQ ID NO: 74; and iii) a third vector comprising a nucleic acid encoding a third polypeptide chain comprising the amino acid sequence of SEQ ID NO: 78. 15. A host cell comprising the vector set of claim 14 . 16. A bispecific binding agent comprising a first binding domain that binds to human CLDN18.2, a second binding domain that binds to human CLDN18.2, and a third binding domain that binds to human CD3, wherein the bispecific binding agent comprises four polypeptide chains, wherein (i) the first polypeptide chain is encoded by a first nucleic acid sequence encoding the amino acid sequence of SEQ ID NO: 73; (ii) the second polypeptide chain is encoded by a second nucleic acid encoding the amino acid sequence of SEQ ID NO: 74; (iii) the third polypeptide chain is encoded by a third nucleic acid encoding the amino acid sequence of SEQ ID NO: 78; and (iv) the fourth polypeptide chain is encoded by the third nucleic acid encoding the amino acid sequence of SEQ ID NO: 78. 17. The bispecific binding agent of claim 16 , wherein the first polypeptide chain comprises the amino acid sequence of SEQ ID NO: 73 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 73 comprises deletion of lysine at position 447 of SEQ ID NO: 73. 18. The bispecific binding agent of claim 16 , wherein the second polypeptide chain comprises the amino acid sequence of SEQ ID NO: 74 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 74 comprises deletion of lysine at position 720 of SEQ ID NO: 74. 19. The bispecific binding agent of claim 16 , wherein the third polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78. 20. The bispecific binding agent of claim 16 , wherein the fourth polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78. 21. A bispecific binding agent comprising a first binding domain that binds to human CLDN18.2, a second binding domain that binds to human CLDN18.2, and a third binding domain that binds to human CD3, wherein the bispecific binding agent comprises four polypeptide chains, i) wherein the first polypeptide chain comprises the amino acid sequence of SEQ ID NO: 73 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 73 comprises deletion of lysine at position 447 of SEQ ID NO: 73; ii) wherein the second polypeptide chain comprises the amino acid sequence of SEQ ID NO: 74 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 74 comprises deletion of lysine at position 720 of SEQ ID NO: 74; iii) wherein the third polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78; and iv) wherein the fourth polypeptide chain comprises the amino acid sequence of SEQ ID NO: 78. 22. A bispecific binding agent comprising a first binding domain that binds to human CLDN18.2, a second binding domain that binds to human CLDN18.2, and a third binding domain that binds to human CD3, wherein the bispecific binding agent comprises four polypeptide chains, i) wherein the first polypeptide chain consists of the amino acid sequence of SEQ ID NO: 73 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 73 comprises deletion of lysine at position 447 of SEQ ID NO: 73; ii) wherein the second polypeptide chain consists of the amino acid sequence of SEQ ID NO: 74 or a C-terminal truncation variant thereof, wherein the C-terminal truncation variant of SEQ ID NO: 74 comprises deletion of lysine at position 720 of SEQ ID NO: 74; iii) wherein the third polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78; and iv) wherein the fourth polypeptide chain consists of the amino acid sequence of SEQ ID NO: 78.
