Described herein are polymerase variants that are exonuclease deficient. Some variants retain the strand displacement capability comparable to the wild-type or parental polymerase. Some variants have a strand displacement capability that is improved relative to the wild-type or parental polymerase. The variants may have an extension rate that is greater than the wild-type or parental polymerase. The variants may have a waiting time that is less than the wild-type or parental polymerase.

What is claimed is: 1. A DNA polymerase comprising an amino acid sequence having at least 80% sequence identity to SEQ ID NO: 1, wherein the DNA polymerase: (A) has a lower 3′→5′ exonuclease activity than a reference DNA polymerase, wherein the reference DNA polymerase consists of SEQ ID NO: 1, (B) has the same or higher strand displacement activity than the reference DNA polymerase, and (C) has a set of substitutions relative to SEQ ID NO: 1, and wherein at least one substitution of the set of substitutions is at a position selected from the group consisting of L125K, D128H, V179Y, V179R, S211F, Y212K, I215L, T216K, Q221R, Y242G/A/L/S, Y259G/K/Q, A292K, S293G, and S294T. 2. The DNA polymerase of claim 1 , wherein the set of substitutions comprises each of T529M, S366A, A547F, and Y242G/A/L/S. 3. The DNA polymerase of claim 1 , wherein the DNA polymerase further has an extension rate that is greater than or equal to the extension rate of the reference DNA polymerase. 4. The DNA polymerase of claim 3 , wherein the extension rate is at least 1.5× greater than the extension rate of the reference DNA polymerase. 5. The DNA polymerase of claim 3 , wherein the extension rate is from 1.5× to 5× greater than the extension rate of the reference DNA polymerase.
