The present disclosure is directed to human monoclonal IgE antibodies, and IgG antibodies engineered therefrom. Such engineered antibodies can be used to blunt pathologic IgE responses in subjects, such as in the treatment or prevention of allergies.

What is claimed is: 1. A pharmaceutical composition comprising: a therapeutically effective amount of an antibody or a fragment thereof, wherein the antibody or the fragment thereof binds a peanut Ara h2 allergen, wherein the antibody or antibody fragment comprises heavy chain CDR1 SEQ ID NO: 108, heavy chain CDR2 SEQ ID NO: 109, heavy chain CDR3 SEQ ID NO: 110, light chain CDR1 SEQ ID NO: 186, light chain CDR2 SEQ ID NO: 187 and light chain CDR3 SEQ ID NO: 188; and a pharmaceutically acceptable carrier. 2. The composition of claim 1 , wherein the antibody is a human IgE mAb. 3. The composition of claim 1 , wherein the antibody or antibody fragment comprises a heavy chain variable region having 90% or greater sequence identity to SEQ ID NO: 55 and a light chain variable region having 90% or more sequence identity to SEQ ID NO: 56, but retaining CDR1 SEQ ID NO: 108, heavy chain CDR2 SEQ ID NO: 109, heavy chain CDR3 SEQ ID NO: 110, light chain CDR1 SEQ ID NO: 186, light chain CDR2 SEQ ID NO: 187 and light chain CDR3 SEQ ID NO: 188. 4. The composition of claim 1 , wherein the antibody or antibody fragment comprises a heavy chain variable region comprising SEQ ID NO: 55 and a light chain variable region comprising SEQ ID NO: 56. 5. The composition of claim 1 wherein the antibody comprises a heavy chain variable region having 80% sequence identity to SEQ ID NO: 55 and a light chain variable region having 80% sequence identity to SEQ ID NO: 56. 6. The composition of claim 1 , wherein the antibody comprises a recombinant single chain fragment variable (scFv) antibody, an Fab fragment, an F(ab′)2 fragment, or an Fv fragment. 7. The composition of claim 1 , wherein the antibody comprises an IgE, or is an IgG comprising grafted IgE CDRs or variable regions. 8. The composition of claim 1 , wherein the antibody further comprises a cell penetrating peptide.
