In various embodiments, compositions and methods are provided for treatment and/or prevention of amyloidogenic diseases. In certain embodiments, the methods entail administering an effective amount of a tropinol ester to a subject in need thereof for prophylactic or therapeutic effect. The methods are particularly useful for prophylactic and therapeutic treatment of Alzheimer's disease. In certain embodiments, methods of reducing the risk, lessening the severity, or delaying the progression or onset of a disease characterized by beta-amyloid deposits in the brain of a mammal are also provided. In certain embodiments, methods of directly or indirectly inhibiting the C-terminal cleavage of APP resulting in the formation of APP-C31 peptide and APPneo (AP-P664) in a mammal are provided.

What is claimed is: 1. A method of treating Alzheimer's disease in a subject in need thereof with one or more risk factors for Alzheimer's disease, said method comprising: administering to said subject a compound selected from the group consisting of or a pharmaceutically acceptable salt, solvate, or hydrate thereof; wherein said one or more risk factors are selected from the group consisting of a) said subject is a carrier of one or more apolipoprotein E (APOE) ε4 alleles and is AD-P biomarker positive, b) said subject is a carrier of an autosomal dominant mutation in one or more of APP, PS1, or PS2, and c) a combination thereof. 2. The method of claim 1 , wherein said method produces an increase in the CSF of said subject of sAPPα. 7. A method of treating Alzheimer's disease in a subject, said method comprising: administering to the subject an amount of 1-1000 mg of a compound selected from the group consisting of or a pharmaceutically acceptable salt, solvate, or hydrate thereof.
