Methods and compositions are provided for enhancing dentin production, and for delivering a lipophilic agent to pulp tissue of a tooth of an individual. In some cases, a subject method includes a step of administering to the pulp of a tooth of an individual, a Wnt stimulating composition that includes a Wnt stimulator agent, at a dose sufficient to enhance the production of dentin by the pulp. In some cases, a subject method includes a step of contacting exposed dentin of a tooth with a composition that includes a lipophilic agent inserted in the non-aqueous phase of a lipid structure (e.g., whereby the lipophilic agent penetrates the dentin to the underlying pulp tissue). Kits are also provided for practicing the methods of the disclosure.

What is claimed: 1. A method for enhancing dentin production, the method comprising: (a) providing a tissue from a subject; (b) contacting the tissue with liposomal Wnt3A, wherein the liposomal Wnt3A comprises a Wnt3A protein inserted into a non-aqueous phase of a lipid structure, thereby forming a Wnt3A-modified tissue; and (c) administering the Wnt3A-modified tissue to a tooth of the subject. 2. The method according to claim 1 , wherein the Wnt3A protein is human Wnt3A. 3. The method according to claim 1 , wherein the Wnt3A protein has an amino acid sequence of SEQ ID NO: 19. 4. The method according to claim 1 , wherein the lipid structure comprises a size range of from 0.03 to 0.2 micron. 5. The method according to claim 1 , wherein the tissue is derived from dental pulp. 6. The method according to claim 1 , wherein the tissue is derived from bone marrow. 7. The method according to claim 1 , wherein the tissue provided from the subject is from the tooth to which the Wnt3A-modified tissue is administered. 8. The method according to claim 1 , wherein the tissue provided from the subject is from a different tooth of the subject than that to which the Wnt3A-modified tissue is administered. 9. The method according to claim 1 , wherein the Wnt3A-modified tissue is present in an amount sufficient to activate stem cells, progenitor cells, or a combination thereof in the tissue. 10. The method according to claim 1 , wherein the Wnt3A-modified tissue is present in an amount sufficient to reduce apoptosis of a dental pulp tissue in the tooth of the subject. 11. The method according to claim 1 , wherein the Wnt3A-modified tissue is administered to dental pulp tissue within the tooth. 12. The method according to claim 1 , wherein administering the Wnt3A-modified tissue increases the number of differentiated odontoblasts in the tooth. 13. The method according to claim 1 , wherein the Wnt3A-modified tissue promotes secretion of dentin matrix from odontoblasts in the tooth. 14. The method according to claim 1 , wherein the Wnt3A-modified tissue further comprises a pharmaceutically acceptable carrier. 15. The method according to claim 1 , wherein the Wnt3A-modified tissue is administered as a single administration. 16. The method according to claim 1 , wherein the Wnt3A-modified tissue is administered as part of a series of administrations. 17. The method according to claim 1 , wherein the subject is a human. 18. The method according to claim 1 , wherein the method is performed as part of a dental procedure. 19. The method according to claim 18 , wherein the dental procedure is a deep dental restoration. 20. The method according to claim 19 , wherein the deep dental restoration comprises placement of an amalgam, composite, or crown within the tooth.
