Streptococcus pneumoniae is a major health concern, especially in very young, elderly, or immunocompromised patients. The present disclosure provides, inter alia, certain highly effective vaccines and pharmaceutical compositions in Streptococcus pneumoniae. The antigens may be used therapeutically or prophylactically.

First claim

We claim: 1. A vaccine formulation comprising: (i) a pharmaceutically acceptable carrier, (ii) an effective amount of an adjuvant, (iii) a first polypeptide comprising an amino acid sequence at least 95% identical to SEQ ID 6 or SEQ ID 7, and (iv) a second polypeptide comprising an amino acid sequence at least 95% identical to SEQ ID 9 or SEQ ID 10. 2. The vaccine formulation of claim 1 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 9. 3. The vaccine formulation of claim 1 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 10. 4. The vaccine formulation of claim 1 , wherein the polypeptide is conjugated to an immunogenic carrier. 5. The vaccine formulation of claim 1 , which comprises at least one lipidated polypeptide. 6. The vaccine formulation of claim 1 , wherein the adjuvant is an agonist of toll-like receptors (TLRs). 7. The vaccine formulation of claim 1 , wherein the adjuvant is alum. 8. The vaccine formulation of claim 1 , wherein the vaccine formulation inhibits infection by Streptococcus pneumoniae in an uninfected subject. 9. The vaccine formulation of claim 1 , wherein the vaccine formulation inhibits Streptococcus pneumoniae colonization in an individual. 10. The vaccine formulation of claim 1 , wherein the vaccine formulation inhibits Streptococcus pneumoniae symptoms. 11. A method for treating a subject suffering from or susceptible to Streptococcus pneumoniae infection, comprising administering an effective amount of a vaccine formulation according to claim 1 . 12. The method of claim 11 , wherein the method inhibits infection by Streptococcus pneumoniae in an uninfected subject. 13. The method of claim 11 , wherein the method inhibits Streptococcus pneumoniae colonization in an individual. 14. The method of claim 11 , wherein the method inhibits Streptococcus pneumoniae symptoms. 15. The method of claim 11 , wherein the method treats a subject with one dose. 16. The method of claim 11 , wherein the method treats a subject within three doses. 17. The method of claim 11 , wherein the subject is a human. 18. The vaccine formulation of claim 1 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 6. 19. The vaccine formulation of claim 1 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 7. 20. A pharmaceutical composition comprising: (i) a pharmaceutically acceptable carrier, (ii) an effective amount of an adjuvant, (iii) a first polypeptide comprising an amino acid sequence at least 95% identical to SEQ ID 6 or SEQ ID 7, and (iv) a second polypeptide comprising an amino acid sequence at least 95% identical to SEQ ID 9 or SEQ ID 10. 21. The pharmaceutical composition of claim 20 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 6. 22. The pharmaceutical composition of claim 20 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 7. 23. The pharmaceutical composition of claim 20 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 9. 24. The pharmaceutical composition of claim 20 , which comprises a polypeptide having an amino acid sequence comprising SEQ ID NO: 10. 25. The pharmaceutical composition of claim 20 , which comprises a polypeptide having an amino acid sequence consisting of SEQ ID NO: 6. 26.
