Disclosed herein are monospecific HCAb antibodies with antigen-binding specificity to ANG-2 and bispecific antibodies with antigen-binding specificities to ANG-2 and VEGF or PDGF.

What is claimed is: 1. A method of treating age-related macular degeneration comprising administering to a subject in need thereof a heavy chain antibody (HCAb) with an antigen-binding specificity for ANG-2, wherein the HCAb comprises three complementarity determining regions (CDR), wherein: the first CDR (CDR1) comprises SEQ ID NO:12; the second CDR (CDR2) comprises SEQ ID NO:14; and the third CDR (CDR3) comprises one of SEQ ID NOs:16, 28, or 31. 2. The method of claim 1 , wherein CDR3 comprises SEQ ID NO. 16. 3. The method of claim 1 , wherein CDR3 comprises SEQ ID NO:28. 4. The method of claim 1 , wherein CDR3 comprises SEQ ID NO:31. 5. A method of treating age-related macular degeneration comprising administering to a subject in need thereof a heavy chain antibody (HCAb) with an antigen-binding specificity for ANG-2, wherein the HCAb comprises three CDRs, wherein CDR1 comprises SEQ ID NO:32, CDR2 comprises SEQ ID NO:33, and CDR3 comprises SEQ ID NO:34. 6. A method of treating age-related macular degeneration comprising administering to a subject in need thereof a heavy chain antibody (HCAb) with an antigen-binding specificity for ANG-2, wherein the HCAb comprises three CDRs, wherein CDR1 comprises SEQ ID NO:35, CDR2 comprises SEQ ID NO:36, and CDR3 comprises SEQ ID NO:37. 7. The method of claim 1 , wherein the age-related macular degeneration is wet age-related macular degeneration or neovascular age-related macular degeneration. 8. The method of claim 7 , wherein the wet age-related macular degeneration or the neovascular age-related macular degeneration is associated with aging. 9. The method of claim 7 , wherein the wet age-related macular degeneration or the neovascular age-related macular degeneration is caused by severe myopia. 10. The method of claim 7 , wherein the wet age-related macular degeneration or the neovascular age-related macular degeneration is caused by intraocular infections such as histoplasmosis. 11. The method of claim 1 , wherein the age-related macular degeneration is dry age-related macular degeneration or non-neovascular age-related macular degeneration. 12. The method of claim 11 , wherein the dry age-related macular degeneration or the non-neovascular age-related macular degeneration is caused by distortion, pigmentation, or thinning of the retina. 13. The method of claim 1 , wherein the heavy chain antibody (HCAb) is administered in a sustained release formulation. 14. The method of claim 13 , wherein the heavy chain antibody (HCAb) is released into the vitreous over a 3-6-month period.
