Abstract

Official abstract text for this publication.

A carrier for holding a biological sample includes a substrate. The substrate is configured to engage a first sample chamber comprising a first opening characterized by a first opening diameter or a second sample chamber comprising a second opening characterized by a second opening diameter that is greater than the first opening diameter. The substrate includes an upper portion, a lower portion, and an intermediate portion disposed between the upper portion and the lower portion. The lower portion is disposed below the upper portion and comprises a bottom surface configured to receive a biological sample. The intermediate portion is characterized by a first substrate diameter and the lower portion is characterized by a second substrate diameter that is less than the first substrate diameter.

First claim

What is claimed is: 1. A method for processing a biological sample comprising: capturing a cell sample on a transfer film disposed on a bottom surface of a sample carrier cap, wherein the sample carrier cap is configured with a first seating area and a second seating area for interference fit with a variety of sample chamber opening diameters; selecting a sample chamber having an opening diameter dimensioned to engage with the first or the second seating area of the sample carrier cap and a volume to accommodate processing the cell sample; engaging the sample carrier cap with the opening of the sample chamber to provide an interference fit, wherein the sample carrier cap is configured to reversibly seal the sample chamber during use; and processing the cell sample to provide a cell lysate in the sample chamber. 2. The method of claim 1 , further comprising performing an assay on the cell lysate. 3. The method of claim 2 , wherein performing an assay is performing a sequencing assay on the cell lysate. 4. The method of claim 2 , wherein performing an assay is performing a polymerase chain reaction (PCR) assay on the cell lysate. 5. The method of claim 4 , wherein the PCR assay performed is a quantitative PCR assay. 6. The method of claim 1 , further comprising: transferring the cell lysate to a filter column, wherein the filter column is configured to extract nucleic acid species from the lysate; and eluting the extracted nucleic acid species from the filter column into a reaction tube. 7. The method of claim 6 , further comprising performing an assay on the extracted nucleic acid species. 8. The method of claim 7 , wherein performing an assay is performing a sequencing assay on the extracted nucleic acid species. 9. The method of claim 7 , wherein performing an assay is performing a polymerase chain reaction (PCR) assay on the extracted nucleic acid species. 10. The method of claim 9 , wherein the PCR assay is a quantitative PCR assay. 11. The method of claim 6 , wherein eluting the extracted nucleic acid species from the filter column comprises eluting the extracted nucleic acid species in a smaller volume than a lysis volume used in processing the cell sample, thereby concentrating the extracted nucleic acid species. 12. The method of claim 1 , wherein the sample chamber selected has a volume of between 0.10 milliliters to 2.0 milliliters. 13. The method of claim 12 , wherein the sample chamber is a 0.20 milliliter tube. 14. The method of claim 12 , wherein the sample chamber is a 0.50 milliliter tube. 15. The method of claim 1 , wherein engaging the sample carrier cap with the opening of the sample chamber provides an O-ring seal between the sample carrier cap and the sample chamber.
