The present disclosure provides antibodies and antigen-binding fragments capable of binding PrP C . The disclosure further provides methods for making and using the antibodies and antigen-binding fragments.

First claim

We claim: 1. An antibody or antigen-binding fragment, wherein the antibody or antigen-binding fragment comprises a VH CDR1 having the amino acid sequence of SEQ ID NO: 5, a VH CDR2 having the amino acid sequence of SEQ ID NO: 6, and a VH CDR3 having the amino acid sequence of SEQ ID NO: 7, and comprises a VL CDR1 having the amino acid sequence of SEQ ID NO: 14, a VL CDR2 having the amino acid sequence of SEQ ID NO: 15, and a VL CDR3 having the amino acid sequence of SEQ ID NO: 16. 2. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a light chain variable (VL) domain and a heavy chain variable (VH) domain; wherein the VL domain comprises CDR1, CDR2 and CDR3 of the amino acid sequence set forth in SEQ ID NO: 13. 3. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment comprises a light chain variable (VL) domain and a heavy chain variable (VH) domain; wherein the VH domain comprises CDR1, CDR2 and CDR3 of the amino acid sequence set forth in SEQ ID NO: 4. 4. The antibody or antigen-binding fragment of claim 1 , wherein the VH domain comprises framework regions that are at least 90% identical to the amino acid sequences of one or more of SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 10 and SEQ ID NO: 11. 5. The antibody or antigen-binding fragment of claim 1 , wherein the VL domain comprises framework regions that are at least 90% identical to the amino acid sequences of one or more of SEQ ID NO: 17, SEQ ID NO: 18, SEQ ID NO: 19 and SEQ ID NO: 20. 6. The antibody or antigen-binding fragment of claim 1 , wherein the VH domain comprises an amino acid sequence that is at least 90% identical to SEQ ID NO: 4 and/or the VL domain comprises an amino acid sequence that is at least 90% identical to SEQ ID NO: 13. 7. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment is humanized or human. 8. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment is encoded by a nucleic acid comprising a nucleotide sequence that is at least 90% identical to SEQ ID NO: 3 or a nucleotide sequence that is at least 90% identical to SEQ ID NO: 12. 9. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment competes with 6D11 antibody for binding to PrP c . 10. The antibody or antigen-binding fragment of claim 1 , wherein the antibody or antigen-binding fragment binds to one or more of human, rat, mouse, or cynomolgus monkey PrP c . 11. A composition comprising a pharmaceutically acceptable carrier and the antibody or antigen-binding fragment of claim 1 . 12. A kit comprising the antibody or antigen-binding fragment of claim 1 .
