Described are dimethoxyphenyl inhibitors of VMAT2, pharmaceutical compositions thereof, and methods of use thereof.

What is claimed is: 1. A compound of Formula I: or a salt thereof, wherein: R 1 -R 2 and R 21 are independently —CH 3 , —CH 2 D, —CD 2 H, or —CD 3 ; and (i) R 6 and R 7 are deuterium; and R 3 -R 5 and R 8 -R 20 are, independently, hydrogen or deuterium; or (ii) R 8 and R 9 are deuterium; and R 3 -R 7 and R 10 -R 20 are, independently, hydrogen or deuterium; or (iii) R 6 -R 9 are deuterium; and R 3 -R 5 and R 10 -R 20 are, independently, hydrogen or deuterium; or (iv) R 11 and R 12 are deuterium; and R 3 -R 10 and R 13 -R 20 are, independently, hydrogen or deuterium; or (v) R 13 is deuterium; and R 3 -R 12 and R 14 -R 20 are, independently, hydrogen or deuterium; or (vi) R 15 and R 16 are deuterium; and R 3 -R 14 and R 17 -R 20 are, independently, hydrogen or deuterium; and at least one of the deuteriums has a deuterium enrichment of no less than about 10%. 2. The compound as recited in claim 1 wherein R 10 and R 14 are hydrogen. 3. The compound as recited in claim 1 wherein R 1 is CD 3 . 4. The compound as recited in claim 1 wherein R 2 is CD 3 . 5. The compound as recited in claim 1 wherein R 21 is CD 3 . 6. The compound as recited in claim 1 wherein R 6 and R 7 are deuterium. 7. The compound as recited in claim 1 wherein R 8 and R 9 are deuterium. 8. The compound as recited in claim 1 wherein R 6 -R 9 are deuterium. 9.
