The present invention relates to isolated polypeptides having cellobiohydrolase activity, catalytic domains, and cellulose binding domains and polynucleotides encoding the polypeptides, catalytic domains, and cellulose binding domains. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides, catalytic domains, or cellulose binding domains.

First claim

What is claimed is: 1. A method for degrading a cellulosic material, comprising: treating the cellulosic material with an enzyme composition comprising a polypeptide having cellobiohydrolase activity, wherein the polypeptide having cellobiohydrolase activity comprises an amino acid sequence having at least 95% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 2. The method of claim 1 , further comprising recovering the degraded cellulosic material. 3. A method for producing a fermentation product, comprising: (a) saccharifying a cellulosic material with an enzyme composition comprising a polypeptide having cellobiohydrolase activity, wherein the polypeptide having cellobiohydrolase activity comprises an amino acid sequence having at least 95% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2; (b) fermenting the saccarified cellulosic material with one or more fermenting microorganisms to produce the fermentation product; and (c) recovering the fermentation product from the fermentation. 4. A method of fermenting a cellulosic material, comprising: fermenting the cellulosic material with one or more fermenting microorganisms, wherein the cellulosic material is saccharified with an enzyme composition comprising a polypeptide having cellobiohydrolase activity, wherein the polypeptide having cellobiohydrolase activity comprises an amino acid sequence having at least 95% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 5. The method of claim 4 , wherein the fermenting of the cellulosic material produces a fermentation product. 6. The method of claim 5 , further comprising recovering the fermentation product from the fermentation. 7. A whole broth formulation or cell culture composition comprising a polypeptide having cellobiohydralase activity, wherein the polypeptide having cellobiohydralase activity comprises an amino acid sequence having at least 95% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 8. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 96% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 9. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 97% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 10. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 98% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 11. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity has at least 99% sequence identity to amino acids 20 to 456 of SEQ ID NO: 2. 12. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity comprises amino acids 20 to 456 of SEQ ID NO: 2. 13. The method of claim 1 , wherein the polypeptide having cellobiohydrolase activity is encoded by the polynucleotide of SEQ ID NO: 1 which is identical to the polynucleotide contained in plasmid pAJ227 which is contained in E. coli deposited with the Agricultural Research Service Patent Culture Collection (NRRL) under accession number B-50474. 14. The method of claim 1 , wherein the cellulosic material is pretreated. 15. The method of claim 1 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 16. The method of claim 2 , wherein the degraded cellulosic material is a sugar. 17. The method of claim 16 , wherein the sugar is selected from the group consisting of glucose, xylose, mannose, galactose, and arabinose. 18. The method of claim 3 , wherein the polypeptide having cellobiohydrolase activity comprises amino acids 20 to 456 of SEQ ID NO: 2. 19. The method of claim 3 , wherein the cellulosic material is pretreated. 20. The method of claim 3 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 21. The method of claim 3 , wherein steps (a) and (b) are performed simultaneously in a simultaneous saccharification and fermentation. 22. The method of claim 3 , wherein the fermentation product is an alcohol, an alkane, a cycloalkane, an alkene, an amino acid, a gas, isoprene, a ketone, an organic acid, or polyketide. 23. The method of claim 4 , wherein the polypeptide having cellobiohydrolase activity comprises amino acids 20 to 456 of SEQ ID NO: 2. 24. The method of claim 4 , wherein the cellulosic material is pretreated. 25. The method of claim 4 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 26. The method of claim 5 , wherein the fermentation product is an alcohol, an alkane, a cycloalkane, an alkene, an amino acid, a gas, isoprene, a ketone, an organic acid, or polyketide.
